The European Union Clinical Trials Register   allows you to search for protocol and results information on:
  • interventional clinical trials that were approved in the European Union (EU)/European Economic Area (EEA) under the Clinical Trials Directive 2001/20/EC
  • clinical trials conducted outside the EU/EEA that are linked to European paediatric-medicine development

  • EU/EEA interventional clinical trials approved under or transitioned to the Clinical Trial Regulation 536/2014 are publicly accessible through the
    Clinical Trials Information System (CTIS).


    The EU Clinical Trials Register currently displays   43857   clinical trials with a EudraCT protocol, of which   7284   are clinical trials conducted with subjects less than 18 years old.   The register also displays information on   18700   older paediatric trials (in scope of Article 45 of the Paediatric Regulation (EC) No 1901/2006).

    Phase 1 trials conducted solely on adults and that are not part of an agreed paediatric investigation plan (PIP) are not publicly available (see Frequently Asked Questions ).

    E.2 Objective of the trial
    E.2.1Main objective of the trial
    To evaluate simultaneously the Overall Response Rate (ORR)of oral vinorelbine in combination with capecitabine, gemcitabine in combination with paclitaxel and gemcitabine in combination with docetaxel in HER2 negative metastatic breats cancer patients.
    Obiettivo primario: •Valutare contemporaneamente la percentuale di risposta oggettiva della combinazione orale vinorelbine + capecitabina e delle combinazioni, somministrate per via endovenosa, gemcitabina + paclitaxel e gemcitabina + docetaxel in pazienti HER2 negative con carcinoma mammario metastatico.

    - To assess the safety profile of the 3 arms. - To evaluate other efficacy parameters in each arm: a)progression free survival; b)duration of response; c)time to treatment failure; d)ovrall survival. - To assess the quality of life of the 3 arms.

    E.3Principal inclusion criteria
    Women with: Age =/> 18years; Histologically confirmed adenocarcinoma of the breast; Documented metastatic disease previously untreated with chemoterapy; HER2 negative (assessed by ICH or FISH methods) on the primary tumor or on metastatic site; At least one measurable lesion using the RECIST criteria; Women on childbearing potential must be using a medically accepted method of contraception (oral contraceptives, intrauterine devices)to avoid pregnancy during the 2 months preceding the start of study treatment, throughout the study period and for up to 3 months after the last dose of study treatment in such a manner to minimise the risk of pregnancy; Negative serum or urine pregnancy test within 72 hours prior to the start of study treatment; Prior chemoterapy as follows: -Adjuvant or neoadjuvant chemotherapy wich may contain an anthracycline is allowed and relapsing more than 6 months after the end of chemotherapy. - Patient previously treated with 5-FU infusion in adjuvant setting is allowed and relapsing more than 6 months after the end of chemotherapy; Previous hormonotherapy for metastatic breast cancer is allowed; Patients may have received prior therapy but not on the sites used to assess the response. A minimum of 4 weeks interval must have elapsed; Karnofsky Performance Status =/> 70%; Life expectancy =/> 16 weeks; Adequate bone marrow, hepatic and renal functions as evidenced by the following:-Haemoglobin =/> 10 g/dL. -Absolute Neutrophil Count =/> 2 x10^9/L. - Platelet Count >/= 100 x 10^9/L. - Total Bilirubin < ULN (ULN: Upper Limit of Normal). - SGOT/SGPT >/= 2.5 x ULN. -Alkaline Phosphatase < 2.5 x ULN )or < 5ULN in case of bone metastases). -Creatinine clearance >50mL/min (calculated using the Cockroft and Gault formula); Absence of psychological, familiar, sociological or geographic condition potentially hampering compliance with the study protocol and follow-up schedule;these conditions should be assessed with the patient before registration in the trial; The patient must give written (personally signed and dated) informed consent before completing any study-related procedure.
    Donne con: Eta` 18 anni; Conferma istologica di adenocarcinoma della mammella; Malattia metastatica documentata non precedentemente trattata con chemioterapia; HER2 negativo (verificato con metodo immunoistochimico o FISH) sul tumore primario o sulla metastasi; Almeno una lesione misurabile secondo i criteri RECIST; Donne che se potenzialmente fertili, abbiano usato ed usino un efficace metodo di contraccezione(contraccetivi orali, dispositivi intrauterini) nei 2 mesi precedenti l`inizio dello trattamento, durante tutto il periodo dello studio e nei 3 mesi successivi all`ultima dose del trattamento in modo che il rischio di una gravidanza venga minimmizzato; Test di gravidanza (urine o siero) con risultato negativo nelle 72 ore precedenti l`inizio dello studio; Precedente chemioterapia: `` e` consentita una precedente chemioterapia adiuvante o neoadiuvante con antraciclina purche` la recidiva si manifesti dopo piu` di 6 mesi dalla fine della chemioterapia. `` e` consenito un precedente trattamento con 5 FU in regime adiuvante se la recidiva si evidenzia dopo piu` di 6 mesi dalla fine della chemioterapia. E` consentita una precedente terapia ormonale se fatta in regime (neo)adiuvante o metastatico. Precedente radioterapia, ma non a livello delle lesioni target. Deve essere osservato un intervallo di almeno 4 settimane dalla fine della radioterapia. Karnofsky Performance Status =/&gt; 70% Aspettativa di vita =/&gt; 16 settimane. Adeguate funzioni del midollo osseo, epatiche e renali: - Emooglobina =/&gt; 10 g/dL; - Conta Assoluta dei Neutrofili =/&gt; 2 x 109/L; - Conta delle Piastrine &gt;/= 100 x 10^9/L; - Bilirubina Totale &lt; ULN (ULN: Limite Superiore di Normalita`); - SGOT/SGPT 2.5 x ULN - Fosfatasi Alcalina &lt; 2.5 ULN (o &lt; 5 ULN in caso di metastasi ossee); - Clearance della Creatinina &gt; 50 mL/min; calcolata secondo la formula di Cockroft e Gault. Assenza di impedimenti psicologici, familiari, sociologici o geografici che potrebbero influire con il trattamento e i follow-up previsti. Questi impedimenti dovranno essere discussi con la paziente prima della registrazione nello studio; Assenza di impedimenti psicologici, famigliari, sociologici o geografici che potrebbero influire con il trattamento ed i follow-up previsti.     Local relapse alone or after conservative treatment or contra-lateral tumor; Female is not eligible to enter the study if: -pregnant or lacating; -with positive pregnancy test at inclusion; Female of childbearing potential who is unwilling or unable to use a medically accepted method to avoid pregnancy during the 2 months preceding the start of study treatment, throughout the study period and at least 3 months following the last dose of study treatment; Patient with symptoms suggesting CNS involvement or leptomeningeal metastases; Concomitant hormonal therapy for metastatic breast cancer; Malabsorbtion syndrome or disease significantly affecting gastrointestinal function or major resection of the stomach or proximal small bowel that could affect absorption of capecitabine and oral vinorelbine; Prior severe and unexspected reaction to fluoropyrimidine therapy (with or without documented DPD deficiency) or known hypersensitivity to 5-fluorouracil, or patient with a history of hypersensitivity reactions to product containing chremaphor; Prior chemotherapy in the metastatic setting; Patients previously treated with a vinca alkaloid, capecitabine, gemcitabine or taxanes; Current pheripheral neuropathy >/= G2 according to NCI criteria; Patients with dysphagia or inability to swallow the tablets; Other serious illness or medical conditions: a)Cardiac disease, b)Unstable diabetes, c)Uncotrolled hypercalcemia, d)Clinically significant active infections, e) Previous organ allograft; Requirement for concurrent use of the antiviral agent (sorivudine) or chemically related analogues such a brivudine; Paticipation to another clinical trial with any investigational drug within 30 days prior to registration; Hystory of another malignancy within the past five years except basal cell carcinoma of the skin or carcinoma in situ of the cervix.
    Presenza di sola recidiva locale dopo terapia conservativa o tumore contro-laterale; donne in gravidanza o nel periodo allattamento o con test di gravidanza positivo all`inclusione; pazienti in eta` fertile che non vogliono o non possono usare un metodo contraccettivo approvato per evitare una gravidanza nei due mesi precedenti l`inizio del trattamento, nell`intero periodo dello studio e almeno nei 3 mesi dopo la fine del trattamento; pazienti con evidenza clinica di coinvolgimento del sistema nervoso centrale o di metastasi delle leptomeningi; ormonoterapia concomitante per malattia metastatica; sindrome di malassorbimento o patologie della funzionalita` del tratto gastrointestinale, o resezione dello stomaco o dell`intestino prossimale che potrebbero interferire con l`assorbimento della capecitabina e della vinorelbine orale (Navelbine Oral ); precedente reazione grave ed inattesa a terapia con una fluoropirimidina (con o senza documentata deficienza da diidropirimidina deidrogenasi -DPD) o ipersensibilita` nota al 5-fluorouracile, o pazienti con una storia di reazione di ipersensibilita` a prodotti che contengono cremaphor , precedente chemioterapia per malattia metastatica; pazienti precedentemente trattate con alcaloidi della vinca, capecitabina, gemcitabina o taxani; concomitante neuropatia periferica di grado 2; pazienti con disfagia o incapacita` ad ingoiare capsule; altre patologie o condizioni mediche serie: ¢ Patologie cardiache; ¢ Diabete instabile; ¢ Ipercalcemia non controllata; ¢ Infezioni significative e attive clinicamente; ¢ Precedente trapianto di organo; Necessita` di terapia concomitante con un agente antivirale (sorivudina) o analoghi chimici come la brivudina; participazione a un altro studio clinico con farmaco sperimentale nei 30 giorni precedenti la registrazione; storia di altro carcinoma nei 5 anni precedenti ad eccezione del carcinoma basale della cute o del carcinoma in situ della cervice
