    E.2.1Main objective of the trial
    To evaluate the proportion of patients who achieve RVR with DEB025 600mg QD plus
    RBV 400 mg BID, DEB025 800 mg QD plus RBV 400 mg BID and DEB025 1000 mg QD treatment
    groups, in treatment-naïve chronic hepatitis C genotype 2/3 patients.
    Obiettivo primario: valutare la proporzione dei pazienti con epatite cronica C (CHC) naive genotipo 2
    e 3 che raggiungono la risposta virologica rapida (RVR) nei seguenti gruppi di trattamento
    • DEB025 600 mg QD in associazione a RBV 400 mg BID
    • DEB025 800 mg QD in associazione a RBV 400 mg BID
    • DEB025 1000 mg QD
    E.2.2Secondary objectives of the trial
    The key secondary objective is to evaluate the proportion of patients with RVR on interferon free
    treatment who also achieve SVR 24 in treatment-naïve chronic hepatitis C genotype 2/3 patients

    raggiungono anche la risposta virologica sostenuta a 24 settimane dopo la fine del trattamento (SVR)

    E.3Principal inclusion criteria
    Key inclusion criteria are:
    • Written informed consent prior to any assessment;
    • Males or females aged ≥18 and ≤ 65 years;
    • BMI between ≥ 18 and ≤ 32 kg/m2;
    • Chronic hepatitis C virus infection;
    • Plasma HCV RNA ≥ 10000 IU/ml; no upper limit;
    • Infection with HCV genotype 2 or 3;
    • HCV treatment naïve;
    • Female patients of child bearing potential must have a negative pregnancy test immediately prior
    to first administration of study drug.

    • pazienti con un indice di massa corporea (BMI) tra 18 e 36 kg/m2
    • pazienti con epatite cronica C diagnosticata da due valutazioni distanziate da un intervallo di
    almeno 6 mesi (la diagnosi iniziale puo' essere basata sulla positivita' agli anticorpi per HCV
    oppure sulla valutazione quantitativa o qualitativa dell’HCV RNA). Il secondo test puo' essere
    effettuato allo screening. Inoltre si richiede conferma della cronicizzazione della malattia
    tramite un test di positivita' dell’HCV RNA.
    • Pazienti con documentata istologia e biochimica consistente con diagnosi di epatite cronica C
    compensata
    • pazienti con HCV RNA plasmatico ≥ 10000 IU/ml determinato tramite PCR quantitativa o

    E.4Principal exclusion criteria
    Key exclusion criteria are:
    • Use of other investigational drugs at the time of enrollment, or within 30 days or 5 half-lives of
    enrollment, whichever is longer;
    • Any contraindications to peg-IFNα2a and/or RBV treatment;
    • Active or suspected cancer, or a history of malignancy where the risk of recurrence is ≥ 20% within
    2 years;
    • HBsAg positive or HIV positive;
    • Cirrhosis, decompensated liver disease or complicated portal hypertension;
    • Hepatocellular carcinoma;
    • Pregnant, nursing or lactating women or women of child-bearing potential UNLESS they are using
    two birth control methods;
    • Men whose female partners are pregnant or contemplating pregnancy. Fertile males UNLESS the
    patient and his partner agree to comply with acceptable contraception;
    • Ongoing or recent use of any other medication (including over the counter medication and herbal
    products) within 2 weeks before study start or within 5 drug half-lives of that medication (whichever is longer) that are known inhibitors/inducers of cytochrome 450 3A, substrates of cytochrome 450
    3A, substrates of P-gp, or substrates/inhibitors of OATPs, MRP2 or BSEP;
    • Any other cause of relevant liver disease other than HCV;
    • History of moderate, severe, or uncontrolled psychiatric disease, especially depression.
    • Pazienti che stanno assumendo altri farmaci sperimentali al momento dell’arruolamento o che
    li hanno assunti nei 30 giorni precedenti l’inizio dello studio o in un periodo di tempo pari a 5
    emivite del farmaco, considerando il periodo piu' lungo
    • Presenza di malattia neoplastica attiva o sospetta o anamnesi positiva per malattia
    neoplastica in una situazione in cui il rischio di recidiva e' ≥ 20% entro 2 anni
    • pazienti con positivita' per HBsAg e HIV
    • Evidenza di cirrosi al momento dello screening. La valutazione per escludere la presenza di
    cirrosi deve riferirsi ad un periodo di tempo compreso nei 2 anni (biopsia epatica) o nei 6 mesi (elastografia transiente o Fibrotest) precedenti lo screening. Deve essere preso in
    considerazione anche qualsiasi altro segno di cirrosi (es, evidenza di ipertensione portale)
    • Evidenza di carcinoma epatocellulare
    • Donne in gravidanza o allattamento
    • Pazienti che stanno assumendo o hanno recentemente assunto un qualsiasi farmaco
    (compresi i farmaci da banco e i prodotti a base di erbe) nelle 2 settimane precedenti l’inizio
    dello studio o in un periodo di tempo pari a 5 emivite del farmaco (considerando il periodo piu'
    lungo) indicato nella lista dei medicinali non autorizzati e noto per essere:
    􀂃 un inibitore/induttore del citocromo 450 3A,
    􀂃 substrato del citocromo 450 3A,
    􀂃 substrato della glicoproteina P di membrana (P-glycoprotein, P-gp)
    􀂃 substrato/inibitore di trasportatori anionici organici (organic anion-transporting
    polypeptide OATPs),
    􀂃 substrato/inibitore della multidrug resistance protein-2 (MRP2)
    􀂃 substrato/inibitore della pompa di efflusso dei sali biliari (Bile Salt Export
    Pump, BSEP)
    • Conta dei neutrofili &lt; 1500/μl; Hb &lt; 12 g/dl per le donne e &lt; 13 g/dl per gli uomini; piastrine &lt;
    90000/μl; conta delle cellule bianche del sangue &lt; 3000/mm3
    • ALT ≥ 10 volte ULN
    • Clearance della creatinina &lt; 50 ml/min
    • Anamnesi positiva di osteopenia
    • TSH fuori dai limiti dinormalita'; in caso di aumento lieve del TSH (&lt;1,5 volte ULN) i pazienti
    possono essere inclusi se gli ormoni tiroidei T3 e T4 liberi sono entro i limiti di normalita'
