    Evaluation of two doses of two investigational rMenB(+OMV)+MenACWY combination vaccine formulations given at a 0, 2 month schedule to demonstrate that the safety and immunogenicity of the combination vaccine are at least comparable to that of the licensed vaccine, Menveo and to that of rMenB+OMV alone. In addition, this study will provide estimates of the immune response rates to the MenB components of the combination vaccines in order to enable a more appropriate design of Phase 3 studies

    E.2 Objective of the trial
    E.2.1Main objective of the trial
    To demonstrate immunologic non-inferiority of two doses of two different formulations of rMenB with OMV + MenACWY to a single dose of Menveo, as measured by the percentage of subjects with hSBA seroresponse against N. meningitidis serogroups A, C, W-135 and Y, at 30 days after the last vaccination, in healthy adolescent and young adults aged 10 - 25 years.
    E.2.2Secondary objectives of the trial
    1. To estimate the immunogenicity of two doses of two different formulations of rMenB with OMV + MenACWY as measured by the percentage of subjects with hSBA ≥ 1:5 and hSBA ≥ 1:8 against strains of N.meningitidis serogroup B, at 30 days after the last vaccination
    2. To compare the immunogenicity of two doses of two different formulations of rMenB with OMV + MenACWY with a single dose of Menveo, as measured by hSBA GMTs against N. meningitidis serogroups A, C, W-135 and Y, at 30 days after the last vaccination
    3. To compare the immunogenicity of two doses of two different formulations of rMenB with OMV + MenACWY with two doses of rMenB (+OMV) as measured by the percentage of subjects with hSBA ≥ 1:5, hSBA ≥ 1:8, four-fold rise and hSBA GMTs against strains of N.meningitidis serogroup B, at 30 days after the last vaccination

    E.3Principal inclusion criteria
    Individuals eligible to be enrolled into this study are male and female subjects that are:
    1. 10 - 25 years of age inclusive at the time of enrollment;
    2. Have given their written informed consent and assent (as applicable) at the time of enrollment, or if the subject is under age 18 at the time of nrollment, the legal parent/guardian of the subject have given their written consent on behalf of the
    subject
    3. Who are available for all the visits scheduled in the study (i.e., not planning to leave the area before the end of the study period);
    4. In good health as determined by the outcome of medical history, physical examination screening/baseline labs and clinical judgment of the Investigator.
    E.4Principal exclusion criteria
    1. History of any meningococcal vaccine administration;
    2. Current or previous, confirmed or suspected disease caused by N. meningitidis;
    3. Household contact with and/or intimate exposure to an individual with any laboratory confirmed N. meningitidis infection within 60 days of enrollment;
    4. Significant acute or chronic infection within the previous 7 days or fever (defined as oral temperature >/=38°C) within the previous 3 days;
    5. Antibiotics within 7 days prior to vaccination or blood draw;
    6. Pregnant or nursing (breastfeeding) mothers;
    7. Females of childbearing age who have not used or do not plan to use acceptable birth control measures, for the duration of the study. Oral, injected or implanted hormonal contraceptive, barrier methods (condom or diaphragm with spermicide), intrauterine device or sexual abstinence are considered acceptable forms of birth control. If sexually active the subject must have been using one of the accepted birth control methods at least two months prior to study entry;
    8. Any serious, chronic, or progressive disease (e.g., neoplasm, diabetes, cardiac disease, hepatic disease, progressive neurological disease or seizure disorder; autoimmune disease, HIV infection or AIDS, blood dyscrasias, bleeding diathesis, signs of cardiac or renal failure, or severe malnutrition);
    9. Known or suspected impairment/alteration of the immune system, immunosuppressive therapy, including use of corticosteroids in immunosuppressive doses or chronic use of inhaled high-potency corticosteroids or immunostimulants within the previous 60 days. Use of topical corticosteroids administered during the study in limited areas (i.e., eczema on knees or face or elbows) of the body is allowed
    10. Receipt of blood, blood products and/or plasma derivatives, or a parenteral immunoglobulin preparation within the previous 90 days;
    11. History of severe allergic reactions after previous vaccinations or hypersensitivity to any vaccine component;
    12. Individuals who received any other licensed vaccines within 2 weeks (for inactivated vaccines) or 4 weeks (for live vaccines) prior to enrollment in this study or who are planning to receive any licensed vaccine within 4 weeks from the study vaccines.
    13. Individuals participating in any clinical trial with another investigational product 30 days prior to first study visit or intent to participate in another clinical study at any time during the conduct of this study;
    14. Individuals who are part of study personnel or close family members conducting this study;
    15. Individuals with medical history or any illness that, in the opinion of the Investigator, might interfere with the results of the study or pose additional risk to the subjects due to participation in the study.

    Immunogenicity
    At all time points, the following will be evaluated:
    1. Percentage of subjects with seroresponse to N. meningitidis serogroups A, C, W-135 and Y
    2. Percentage of subjects with hSBA ≥ 1:8 for N. meningitidis serogroups A, C, W-135 and Y
    3. hSBA ≥ 1:5, hSBA ≥ 1:8 and four-fold rise for N. meningitidis serogroup B strains
    4. Geometric Mean Titers (GMTs) for N. meningitidis serogroups A, C, W-135 and Y and serogroup B strains

    Safety
    There are no formal statistical criteria to assess the safety objectives. The safety data that will be
    collected are:
    1. Local reactions (i.e., pain, erythema, and induration), systemic reactions (i.e., chills, nausea,
    fatigue, myalgia, arthralgia, loss of appetite, headache, rash), fever (body temperature ≥ 38°C), AEs, and SAEs. These safety data will be collected daily for 7 days after each vaccination.
    2. From Day 8 through Day 91 all AEs, SAEs and concomitant medications will be collected in the Diary Card. From Day 92 (31 days following vaccination 2) to Day 241 (180 days following vaccination 2) only medically attended AEs, SAEs, and AEs leading to withdrawal from the study will be collected.
