    The EU Clinical Trials Register currently displays   43876   clinical trials with a EudraCT protocol, of which   7294   are clinical trials conducted with subjects less than 18 years old.   The register also displays information on   18700   older paediatric trials (in scope of Article 45 of the Paediatric Regulation (EC) No 1901/2006).

    Phase 1 trials conducted solely on adults and that are not part of an agreed paediatric investigation plan (PIP) are not publicly available (see Frequently Asked Questions ).

    E.2.1Main objective of the trial
    The primary objective of this clinical trial is to evaluate the influence of different therapeutic strategies during the luteal phase (steroids, recombinant LH, hCG at low doses) in endometrial receptivity, in patients at high risk of ovarian hyperstimulation in whom ovulation induction with aGnRH is decided.
    El objetivo primario del ensayo clínico es evaluar la influencia de diferentes estrategias terapéuticas durante la fase lútea (esteroides, LH recombinante, hCG a bajas dosis) en la receptividad endometrial, de pacientes con alto riesgo de hiperestimulación ovárica en las que se decida inducir la ovulación con aGnRH

    1.To evaluate the influence in hormonal levels of such supporting strategies during the luteal phase.
    2.To evaluate the safety of study medication administered in this patient population.
    3.To obtain a usable protocol in patients at high risk of ovarian hyperstimulation in whom ovulation induction with aGnRH is decided.

    E.3Principal inclusion criteria
    1.Aged between 18 and 34 years old.
    2.Ovule donors admitted in the IVI Oocyte Donor Program - Madrid.
    3.Conventional ovarian stimulation protocol.
    4.Normal XX karyotype.
    5.Body mass index (BMI) between 18 and 25 kg / m2.
    6.Negative serologies for Hepatitis B surface antigens (HBs Ag), antibodies for the Hepatitis C virus (HC Hb) and Human Immunodeficiency Virus (HIV) prior to donation.
    7.Normal cervical cytology in the past year.
    8.General informed consent signed by the patients or their legal representative.

    2.Donante de ovocitos aceptada por el Programa de Donación de IVI Madrid
    3. Protocolo de estimulación ovárica convencional.
    4. Cariotipo XX normal.
    5. Indice de Masa Corporal (IMC) entre 18 y 25 kg/m2.
    6. Serologías negativas para Antígeno de superficie del Virus de la Hepatitis B (Ag HBs) , anticuerpos para el Virus de la Hepatitis C (Ab HC) y Virus de la Imunodeficiencia Humana (VIH) previas a la donación.
    7. Citología cervical normal en el último año.
    8. Consentimiento informado general por escrito de las pacientes o su representante legal

    1.Any clinical condition that counterindicates or prevents being able to carry out an endometrial biopsy.
    2.Hypersensitivity, allergy or intolerance to any of the medications prescribed in each of the treatment groups.
    3.Participation in another clinical trial at the time of donation.
    4.Any serious concomitant illness which, according to investigator´s opinion, may compromise the study or affect patients´ tolerability with regard to this treatment.

    E.5.1Primary end point(s)
    The primary efficacy variable will be the evaluation of endometrial receptivity, measured by endometrial microarrays (ERA) tool in each one of the 4 supportive protocols of the luteal phase, compared with the stardard protocol in which the final oocyte maturation will be induced with 250 mcg of hCG .
    This tool provides a comparison of the genetic expression associated with endometrial receptivity, expressed in two different groups: "R" (Receptive) and "IF" (Implantation Failure).
    These results indicate a high (R) or low (IF) probability of successs with regard to embryonic implantation in the following cycle with ovarian stimulation.
    Variable relacionada con el objetivo primario:
    La variable primaria de eficacia será la valoración de la receptividad endometrial medida con la herramienta de microarrays de endometrio (ERA), en cada uno de los 4 protocolos de soporte de la fase lútea en comparación con el protocolo control estándar en el que la maduración final ovocitaria se inducirá con 250mcg de hCG.
    Dicha herramienta realiza una comparación de la expresión génica asociada a la receptividad endometrial expresada en dos grupos diferentes "R" (Receptivo) o "IF" (Implantation Failure).
    Estos resultados indican una alta (R) o baja (IF) probabilidad de éxito en la implantación embrionaria en el siguiente ciclo con estimulación ovárica.
    E.5.1.1Timepoint(s) of evaluation of this end point
    From the primary endpoint analysis, the percentages of patients in each group with regard to a high or low probability of success concerning embryonic implantation, will be recorded and compared.
    A partir del análisis de la variable principal, se recogerá y compararán los porcentajes de pacientes en cada grupo que presenten alta o baja probabilidad de éxito de implantación embrionaria.
    E.5.2Secondary end point(s)
    Se recogerán las siguientes variables secundarias:
    a) Niveles hormonales:
    Se realizará una evaluación de los niveles de progesterona, estradiol y LH el día de la extracción ovocitaria y su evolución hasta el día LH+7 en el que se realizará la biopsia endometrial en cada uno de los protocolos de soporte de fase lútea.
    b) Seguridad:
    Se valorará la proporción de pacientes que han experimentado acontecimientos adversos.
    The following secondary variables will be recorded:

    a)Hormone levels
    Progesterone, estradiol and LH will be measured the day of the oocyte extraction. The evolution will be followed till LH+7. In this moment, endometrial biopsy will be performed in each of the supportive protocols during the luteal phase.
    B) Safety
    Proportion of patients who have experienced adverse events will be measured.
