    The EU Clinical Trials Register currently displays   43851   clinical trials with a EudraCT protocol, of which   7283   are clinical trials conducted with subjects less than 18 years old.   The register also displays information on   18700   older paediatric trials (in scope of Article 45 of the Paediatric Regulation (EC) No 1901/2006).

    E.2 Objective of the trial
    E.2.1Main objective of the trial
    To demonstrate that the immune response to Hep B antigen of the DTaP-IPV-Hep B-PRP-T is non-inferior to that of the association of PENTAXIM™ + ENGERIX B® 1 month after a three-dose primary series at 2, 3, and 4 months of age.
    E.2.2Secondary objectives of the trial
    Immunogenicity:
    To describe in each group the immunogenicity parameters 1 month after the three-dose primary series at 2, 3, and 4 months of age.

    Safety
    To describe the safety profile (overall and after each vaccine injection) in the two groups, in terms of:
    • Any unsolicited adverse event (AE) in the 30 minutes after each vaccination
    • Any AE and adverse reaction (AR) in the first 7 days after each vaccination
    • Any AEs and ARs in the first 30 days after each vaccination
    • Any serious adverse event (SAE) during the trial (from V01 to 6 months after the last dose of the primary series)

    E.3Principal inclusion criteria
    1) Two-month old infants (from the 50th to the 71st day inclusive) of either gender on the day of inclusion.
    2) Born at full term of pregnancy (≥37 weeks) with a birth weight ≥2.5 kg
    3) Informed Consent Form signed by the parent(s) or other legal representative(s) and an institution official other than an Investigator
    4) Able to attend all scheduled visits and to comply with all trial procedures
    E.4Principal exclusion criteria
    1) Participation in another clinical trial in the 4 weeks preceding the first trial vaccination
    2) Planned participation in another clinical trial during the present trial period
    3) Congenital or acquired immunodeficiency, immunosuppressive therapy such as long-term systemic corticosteroid therapy
    4) Systemic hypersensitivity to any of the vaccine components or history of a life-threatening reaction to a vaccine containing the same substances
    5) Chronic illness at a stage that could interfere with trial conduct or completion
    6) Blood or blood-derived products received since birth
    7) Any vaccination (except Bacillus Calmette-Guerin [BCG]) in the 4 weeks preceding the first trial vaccination
    8) Vaccination planned in the 4 weeks following trial vaccination
    9) History of documented pertussis, T, D, polio, HiB, or Hep B infection(s) (confirmed either clinically, serologically or microbiologically)
    10) Known personal or maternal history of human immunodeficiency virus (HIV), HBsAg, or hepatitis C seropositivity
    11) Previous vaccination against pertussis, T, D, polio, Hib, and Hep B infection(s)
    12) Coagulopathy, thrombocytopenia or a bleeding disorder contraindicating IM
    vaccination
    13) History of seizures
    14) Febrile (axillary temperature 37.4°C [rectal equivalent temperature ≥38.0°C]) or acute illness on the day of inclusion

    Anti-hepatitis B surface antibody titers ≥10 mIU/mL were assessed as the primary immunological endpoint 1 month after the third dose of the primary series (Visit 04 [V04], Day 90 [D90]).

    Immunogenicity (at D90 [V04]):
    The following serological endpoints were assessed 1 month after the third dose of the primary series (V04, D90):
    • Anti-T and anti-D antibody (Ab) titers and seroprotection (defined by titers ≥0.01 IU/mL and ≥0.1 IU/mL)
    • Anti-HBsAb titers at D90 (V04) and seroprotection (defined by titers ≥100 mIU/mL)
    • Anti- polyribosyl ribitol phosphate (PRP) Ab titers and seroprotection (defined by titers ≥0.15 µg/mL, and≥1.0 µg/mL)
    • Anti-pertussis toxoid (PT) and anti-filamentous hemagglutinin (FHA) Ab titers and seroconversion (defined as a 4-fold titer increase from V01 to V04), and the individual V04/V01 titer ratio
    • Anti-polio 1, 2, and 3 Ab titers and seroprotection (defined by titers ≥8 [1/dil])

    Safety/Reactogenicity:
    • Occurrence, nature (Medical Dictionary for Regulatory Activities [MedDRA] preferred term), severity (mild, moderate, severe), and relationship to vaccination for any systemic adverse events (AEs) reported in the 30 minutes after each injection of the investigational or control vaccines
    • Occurrence, time to onset, number of days of occurrence, and severity of solicited (prelisted in the subject diary and case report form [CRF]), injection site reactions, and systemic reactions occurring up to 7 days after each vaccination
    • Occurrence, nature (MedDRA preferred term), time to onset, duration, severity, and relationship to vaccination (for systemic AEs only) of unsolicited (spontaneously reported) AEs up to 30 days after each vaccination
    • Occurrence, nature (MedDRA preferred term), time to onset, duration, severity, and relationship to vaccination of SAEs reported throughout the study (from the first vaccine injection [V01] to 6 months after the last vaccine injection)
