    The EU Clinical Trials Register currently displays   43846   clinical trials with a EudraCT protocol, of which   7282   are clinical trials conducted with subjects less than 18 years old.   The register also displays information on   18700   older paediatric trials (in scope of Article 45 of the Paediatric Regulation (EC) No 1901/2006).

    Phase 1 trials conducted solely on adults and that are not part of an agreed paediatric investigation plan (PIP) are not publicly available (see Frequently Asked Questions ).

    • Tacrolimus whole blood trough level during post-transplant days 1-14 and week 12
    • Total cumulative incidence of inadequate tacrolimus whole blood trough levels during post-transplant days 1-14 (<5 ng/mL or >20 ng/mL)
    • Creatinine serum levels (time frame: post-transplant days 1-14, week 12)
    • Adverse effects of Prograf®/ Advagraf® (time frame: 12 weeks)
    o Increase of serum creatinine >50%
    o Renal failure requiring pharmacological intervention and/ or hemodialysis
    o Neurological effects including encephalopathy, seizures or coma
    • Incidence of acute graft rejection (time frame: 12 weeks)
    • Rejection free time interval (acute cellular rejection) after LT (time frame: 12 weeks)
    • Assessment of graft survival (time frame: 12 weeks)

    - Teilnahme der Patientin/des Patienten an einer anderen klini-schen Prüfung innerhalb der letzten 28 Tage vor dem Ein-schluss
    - Vorhergehende Transplantation eines anderen Organes
    - high urgent-Transplantation, Teillebertransplantation, Lebend-spende-LTX, kombinierte Multiorgantransplantation, AB0-Inkompatibilität
    - Hepatozelluläres Karzinom außerhalb der MILAN-Kriterien, Le-bermetastasen
    - Geplante oder notwendige Zielspiegel von Tacrolimus, die vom Wert 10-15 ng/mL in der ersten Woche abweichen
    - Behandlung oder innerhalb der ersten 14 Tage nach
    Transplantation geplante Behandlung mit den Immunsuppres-siva Ciclosporin A, Sirolimus, Everolimus, Azathioprin.

    - Anzeichen darauf, dass die Patientin/Patient den Prüfplan vo-raussichtlich nicht einhalten wird (z. B. mangelnde Kooperati-onsbereitschaft)
    - bekannte HIV- Infektion
    - Frauen im gebärfähigen Alter, außer Frauen, die eines der folgenden Kriterien erfüllen:
    o negativer Schwangerschaftstest bei Studieneinschluss
    o Post- menopausal (12 Monate natürliche Amenorrhoe)
    o Zustand mindestens 6 Wochen nach Ovarektomie mit oder ohne Hysterektomie

    Der primäre Endpunkt ist der Vergleich der Anzahl (Prozentsatz) derjenigen Patienten, die toxische oder nicht ausreichende Tacrolimus-Talspiegel innerhalb der ersten 14 Tage nach LTX ausweisen. Ein positives Ereignis liegt vor, wenn der Bluttalspiegel von Tacrolimus in den Tagen 2-14 nach LTX zweimalig unterhalb von 5 ng/ mL oder in den Tagen 1-14 nach LTX einmalig überhalb von 20 ng/ mL liegt; andernfalls liegt ein negatives Ereignis vor.

    o Chronische Niereninsuffizienz definiert durch Anstieg des Serum-Kreatinins um mehr als 50% (bis 1 Jahr nach LTX)
    o Zentral neurologische Ereignisse (Encephalopathie, Krämpfanfälle, Koma) bis Tag 14 nach LTX
    o Transplantatabstoßung bis 1 Jahr nach LTX
