    The EU Clinical Trials Register currently displays   43865   clinical trials with a EudraCT protocol, of which   7286   are clinical trials conducted with subjects less than 18 years old.   The register also displays information on   18700   older paediatric trials (in scope of Article 45 of the Paediatric Regulation (EC) No 1901/2006).

    Phase 1 trials conducted solely on adults and that are not part of an agreed paediatric investigation plan (PIP) are not publicly available (see Frequently Asked Questions ).

    E.2.1Main objective of the trial
    1. To evaluate the safety of an intracameral implant of Bimatoprost PF IC DDS in patients with open-angle glaucoma (OAG) and ocular hypertension (OHT) (OHT evaluated in Stage 2 patients only).

    2. To evaluate the intraocular pressure (IOP) lowering effects of an intracameral implant of Bimatoprost PF IC DDS in atients with OAG and OHT (OHT evaluated in Stage 2 patients only)

    E.3Principal inclusion criteria
    Stage 1:
    • Male or female, ≥ 18 years of age
    • Written informed consent and authorization for use and release of personal health information
    • Negative urine pregnancy test results for females of childbearing potential at baseline
    • Patient is a known responder to IOP lowering (minimum of approximately 20% decrease from baseline) by topical prostamides, prostaglandins, or prostaglandin analogues
    • In the study eye, the iridocorneal angle inferiorly must be ≥ grade 3 based on clinical gonioscopic examination. Angle grade must be confirmed as being qualified by Reading Center anterior segment optical coherence tomography (OCT) assessment.
    • In the study eye, patient has diagnosis of OAG (ie, primary,
    pseudoexfoliation, pigmentary) with visual field loss of < -1 db and > -17 db mean deviation at study entry as measured by Humphrey perimetry (24-2 full threshold or Swedish Interactive Thresholding Algorithm [SITA] standard programs). However, this does not include patients with a visual field defect within the central 10 degrees that is, in the opinion of the
    investigator, visually significant or likely to cause central visual impairment upon progression.
    • In the fellow eye, patient has diagnosis of OAG or OHT that can be treated adequately, in the investigator’s opinion, with topical LUMIGAN as sole therapy
    • In both eyes, at baseline visit: hour 0 IOP of ≥ 22 mm Hg and ≤ 36 mm Hg, with difference between eyes of ≤ 3 mm Hg

    Stage 2:
    • Male or female, ≥ 18 years of age
    • Written informed consent and authorization for use and release of personal health information

However, this does not include patients with a visual field defect within the central 10 degrees that is, in the opinion of the investigator, visually significant or likely to cause central visual impairment upon progression; OR
    - OHT requiring IOP-lowering treatment which, in the opinion of the investigator, has a high risk for progression to glaucoma.
    Suggestions to assess this risk include the presence of
    pseudoexfoliation or pigment dispersion syndrome, family history of glaucoma, age, central corneal thickness, and optic nerve head and/or retinal nerve fiber layer analysis. The applicable risk factor(s) should be documented in the patient’s medical record.
    • In the fellow eye, patient has diagnosis of OAG or OHT that can be treated adequately, in investigator’s opinion, with latanoprost or timolol eye drops as the sole therapy
    • In both eyes, at baseline visit: hour 0 IOP of ≥ 22 mm Hg and ≤ 36 mm Hg, with difference between eyes of ≤ 3 mm Hg



    E.4Principal exclusion criteria
    Stage 1 (in either eye):
    - Patients with cataract surgery with posterior capsular tear with or without vitreous loss and/or iris trauma
    - Bulbar conjunctival hyperemia on either macroscopic or slit-lamp examination, > +0.5 (trace) at screening or at baseline
    - History of moderate or worse conjunctival hyperemia due to bimatoprost or other prostaglandin analogs
    - Central endothelial cell counts of < 2000 cells/mm2 by specular microscopy; central corneal thickness of < 470 micrometers or > 630 micrometers, or a 25
    difference between eyes > 70 micrometers
    - History of a narrow angle or closed angle glaucoma
    - Anticipated need for any ocular surgery during the first 12 months of the study period

    Stage 2 (in either eye):
