E.3 | Principal inclusion criteria |
1. Patient with history of chronic obstructive pulmonary disease for at least 12 months prior to screening visit (ATS/ERS 2005) and chronic productive cough for 3 months in each of the 2 years prior to screening visit (if other causes of productive cough have been excluded), and present during the 4 weeks preceding screening. 2. Patient with at least one exacerbation during the preceding year. 3. Patient with FEV1/FVC ratio (post-bronchodilator) ? 70%. 4. Patient with FEV1 (post-bronchodilator) between ? 30 % and ? 50 % of predicted value 5. Patient pre-treated with adequate and stable symptomatic treatment for at least 3 months prior to screening visit. 6. Former smoker (defined as: smoking cessation at least one year ago) or current smoker both with a smoking history of at least 10 pack years 7. Patient with normal organ function defined as: ? absolute neutrophils count (ANC) ? 2.0 x 109/L, ? haemoglobin ? 10 g/dL ? platelets (PTL) ? 100 x 109/L ? AST/ALT ? 3x ULN ? bilirubin ? 1.5x ULN ? creatinin clearance ? 60 mL/min ? albumin > 1 x LLN ? Proteinuria < 30 mg/dL (1+) on the dipstick. If proteinuria is ? 1+ on the dipstick, 24 hours proteinuria must be < 1.5g/24 hours. 8. Male or female patient, aged 40 to 75 years inclusive, weight > 50 kg and BMI between 18 and 35 kg/m² 9. Man and woman of child bearing potential, must agree to use two methods (one for the patient and one for the partner) of medically acceptable forms of contraception during the study and for 3 months after the last treatment intake. 10. Female patient of childbearing potential must have a negative pregnancy test at screening and baseline 11. Patient able and willing to comply with study procedures as per protocol. 12. Patient able to understand the patient card and to follow the patient card procedures in case of signs or symptoms of severe neutropenia or severe cutaneous toxicity, during the first 2 months. 13. Patient able to understand, sign, and date the written voluntary informed consent form at screening visit prior to any protocol-specific procedures. |

Pacientes con al menos una exacerbación durante el año anterior. 3. Pacientes con índice de VEF1/CVF (postbroncodilatador) ? 70%. 4. Pacientes con VEF1 (postbroncodilatador) entre ? 30 % y ? 50 % del valor predictivo. 5. Pacientes que han recibido previamente un tratamiento sintomático apropiado y estable durante al menos los 3 meses anteriores a la visita de selección. 6. Ex fumadores (definidos como: fumadores que dejaron el tabaco al menos un año antes) o fumadores actuales con un índice de consumo de 20 cigarrillos al día durante 10 años en ambos casos. 7. Pacientes con actividad orgánica normal definida como: ? recuento absoluto de neutrófilos (RAN) ? 2,0 x 109/l ? hemoglobina ? 10 g/dl ? plaquetas (Plaq.) ? 100 x 109/l ? AST/ALT ? 3 x LSN ? bilirrubina ? 1,5 x LSN ? aclaramiento de creatinina ? 60 ml/min. ? albúmina > 1 x LIN ? proteinúria < 30 mg/dl (1+) en tira reactiva En caso de proteinuria ? 1+ mg/dl en tira reactiva, proteinuria de 24 horas < 1,5 g/ 24 horas. 8. Pacientes de ambos sexos con edades entre 40 y 75 años inclusive, con un peso > 50 kg y un IMC entre 18 y 35 kg/m². 9. Hombres y mujeres en edad fértil (que participen en el estudio una vez pasado el periodo menstrual y con una prueba de embarazo negativa) deben acceder a utilizar dos métodos anticonceptivos aceptados médicamente (uno para el paciente y el otro para la pareja) durante el estudio y en los 3 meses posteriores a la última toma del tratamiento. 10. Las mujeres en edad fértil deben tener una prueba de embarazo negativa en la visitas de selección y basal. 11. Pacientes capaces y dispuestos a seguir los procedimientos del estudio según el protocolo. 12. Pacientes capaces de entender la tarjeta del paciente y dispuestos a seguir los procedimientos mencionados en ella en caso de observar signos o síntomas de neutropenia grave o toxicidad cutánea grave, durante los primeros 2 meses de tratamiento. 13. Pacientes capaces de entender, firmar y fechar el formulario de consentimiento informado en la visita de selección, antes de que se lleve a cabo cualquier procedimiento específico del protocolo. |

1. Pregnant, or nursing female patient 2. Patient experiencing COPD exacerbation indicated by a treatment with systemic glucocorticosteroids and/or antibiotics not stopped at least 4 weeks prior to the screening visit. 3. Patient with lower respiratory tract infection not resolved 4 weeks prior to the screening visit. 4. Patient with asthma and/or other relevant lung disease (e.g. history of bronchiectasis, cystic fibrosis, bronchiolitis, lung resection, lung cancer, interstitial lung disease [e.g. fibrosis, silicosis, sarcoidosis], and active tuberculosis). 5. Patient currently participating in a pulmonary rehabilitation program or completion of a pulmonary rehabilitation program within 3 months preceding the screening visit. 6. Patients with known alpha-1-antitrypsin deficiency. 7. Patient with clinically significant cardiopulmonary abnormalities (diagnosed clinically or by X-ray / CT-scan / ECG) that are not related to COPD and that require further evaluation. 8. Patient who had major surgery within 2 weeks prior to screening visit 9. Patient presenting with cardiac disorders defined by at least one of the following conditions: ? Patient with recent cardiac history (within 6 months) of: - Acute coronary syndrome - Acute heart failure (class III or IV of the NYHA classification) - Significant ventricular arrhythmia (persistent ventricular tachycardia, ventricular fibrillation, resuscitated sudden death) ? Patient with cardiac failure class III or IV of the NYHA classification ? Patient with severe conduction disorders which are not prevented by permanent pacing (atrio-ventricular block 2 and 3, sino-atrial block) ? Syncope without known aetiology within 3 months ? Uncontrolled severe hypertension, according to the judgment of the investigator, or symptomatic hypertension 10. Patient with: ? Life expectancy < 6 months ? < 5 years free of malignancy, except treated basal cell skin cancer or cervical carcinoma in situ ? Any severe and/or uncontrolled medical condition ? Patient with an active infection (Human immunodeficiency virus infection and/or hepatitis B or C infection, tuberculosis...) 11. Patient with history of poor compliance or history of drug/alcohol abuse, or excessive alcohol beverage consumption that would interfere with the ability to comply with the study protocol, or current or past psychiatric disease that might interfere with the ability to comply with the study protocol or give informed consent 12. 1. Pacientes embarazadas o en periodo de lactancia. 2. Pacientes con exacerbación de EPOC tratada con glucocorticoesteroides sistémicos y/o antibióticos de forma continuada al menos las 4 semanas anteriores a la visita de selección. 3. Pacientes con infección en las vías respiratorias bajas no resuelta 4 semanas antes de la visita de selección. 4. Pacientes con asma y/u otra enfermedad pulmonar relevante (p. ej., antecedentes de broquiectasias, fibrosis quística, bronquiolitis, resección pulmonar, cáncer de pulmón, enfermedad pulmonar intersticial [p. ej., fibrosis, silicosis, sarcoidosis] y tuberculosis activa). 5. Pacientes que estén participando en un programa de rehabilitación pulmonar o que lo hayan finalizado dentro de los 3 meses anteriores a la visita de selección. 6. Pacientes con déficit conocido de alfa-1-antitripsina. 7. Pacientes con anomalías cardiopulmonares clínicamente significativas (diagnosticadas clínicamente o por rayos X / TAC / ECG) que no estén relacionadas con EPOC y que requieran mayor evaluación. 8. Pacientes que se han sometido a cirugía mayor en las 2 semanas anteriores a la visita de selección. 9. Pacientes con afecciones cardíacas definidas por, al menos, uno de los siguientes trastornos: ? Pacientes con antecedentes cardíacos recientes (en los 6 meses previos) de: - Síndrome coronario agudo; - Insuficiencia cardíaca aguda (clase III o IV según la clasificación de la NYHA); - Arritmia ventricular importante (taquicardia ventricular persistente, fibrilación ventricular, muerte súbita reanimada). ? Pacientes con insuficiencia cardíaca de clase III o IV según la clasificación de la NYHA. ? Pacientes con trastornos graves de la conducción cardíaca no evitables mediante electroestimulación cardíaca permanente (bloqueo auriculoventricular 2 y 3, bloqueo sinoauricular). ? Síncope sin etiología conocida en los 3 meses previos. ? Hipertensión grave no controlada, a juicio del investigador, o hipertensión sintomática. 10. Pacientes: ? con una esperanza de vida < 6 meses; ? sin tumor maligno durante < 5 años, excepto carcinoma basocelular o carcinoma cervical localizado tratados; ? con una enfermedad grave y/o no controlada; ? con infección activa (por el virus de inmunodeficiencia humano (VIH) y/o infección por hepatitis B o C, tuberculosis?). 11. Pacientes con antecedentes de falta de cumplimiento terapéutico o drogadicción/alcoholismo, o consumo excesivo de bebidas alcohólicas o enfermedades psiquiátricas presentes o pasadas que pudieran interferir en su capacidad para cumplir el protocolo del estudio o para dar su consentimiento informado. 12.
