    E.2 Objective of the trial
    E.2.1Main objective of the trial
    To evaluate the safety and tolerability of BVS857 in patients with SBMA.
    To evaluate the preliminary efficacy of BVS857 on increasing thigh muscle volume (TMV) in SBMA patients after 12 weeks of dosing (Part B only).
    E.2.2Secondary objectives of the trial
    To assess the effect of BVS857 on extremity muscle function by the Adult Myopathy Assessment Tool (AMAT) in patients with SBMA after 12 weeks of dosing (Part B only).
    To assess the effect of BVS857 on total lean body mass by dual-energy X-ray absorptiometry (DXA) scan in patients with SBMA after 12 weeks of dosing (Part B only).
    Evaluate the pharmacokinetics of BVS857 in SBMA patients, in particular the absolute and relative bioavailability and the dose proportionality.

    •Genetic diagnosis of SBMA with symptomatic muscle weakness
    •Able to complete 2 minute timed walk
    •Serum IGF-1 level less than or equal to 170 ng/mL

    • Medically treated diabetes mellitus or known history of hypoglycemia
    • History of Bell’s palsy
    • Treatment with systemic steroids > 10 mg/day (or equivalent dose);
    androgens or androgen reducing agents; systemic beta agonists; or other muscle anabolic drugs within the previous 3 months
    • History of cancer, other than non-melanomatous skin cancer
    • Retinopathy
    • Papilledema

    Patients will return to the clinic one, four and eight weeks after the final dose of study drug for pharmacokinetic, pharmacodynamic, efficacy and safety assessments. At the final visit (Week 20), patients will undergo Study Completion evaluations and will be discharged from the study.
