E.3 | Principal inclusion criteria |
1) Males and non-pregnant, non-lactating females age = 18 years. 2) Prior successful completion of an avoralstat therapeutic study OR a laboratory confirmed diagnosis of HAE Type 1 (low C1 INH antigenic level) or HAE Type 2 (low C1 INH functional level). 3) Agreement by the Investigator and subject that the subject is a suitable candidate for prophylaxis of HAE attacks. 4) Access to appropriate medication for the treatment of acute HAE attacks. 5) Female subjects must meet one of the following requirements: a) A woman of childbearing potential (defined as a nonmenopausal female who has not had a hysterectomy, bilateral oophorectomy, or documented ovarian failure) who agrees to use a highly effective method of contraception for the study duration and for 30 days after study drug dosing. The following methods are considered to be highly effective methods of contraception: i) surgical sterilization (i.e., bilateral tubal ligation) ii) partner’s surgical sterilization (i.e., vasectomy) iii) placement of an intrauterine device (IUD) or intrauterine system iv) hormonal contraception (implantable, patch, oral, vaginal ring), v) 2 barrier methods (their partner’s use of a condom and the subject’s use of an occlusive cap [diaphragm, or cervical/vault caps] with spermicidal foam/gel/film/ cream/suppository)
Female subjects who report being postmenopausal for = 2 years and have a screening follicle-stimulating hormone (FSH) = 40 mIU/mL must agree to use a highly effective contraceptive method as outlined above for the study period and for 30 days after the last dose of study drug. b) A woman of nonchildbearing potential (defined as being postmenopausal for > 2 years, having a screening FSH > 40 mIU/mL if postmenopausal for = 2 years, or a woman who has had a hysterectomy, bilateral oophorectomy, or documented ovarian failure). c) A woman declaring herself as either sexually abstinent or exclusively having female sexual partners. Abstinence in this study is defined as “True abstinence: when this is in line with the preferred and usual lifestyle of the subject.” All women of childbearing potential making this declaration must consent to utilizing a highly effective method of birth control prior to engaging in any sexual activity with male partners through 30 days after the last dose of study drug. 6) Male subjects must comply with the following requirements: a) Sexually active male subjects must agree to utilize one highly effective contraceptive method with female partners of childbearing potential (defined as postmenopausal = 2 years or a nonmenopausal female who has not had a hysterectomy, bilateral oophorectomy, or documented ovarian failure) through the study period and for 90 days after the last dose of study drug.
A “highly effective contraceptive method” is defined as one of the following: i) subject having had a vasectomy ii) partner’s surgical sterilization (i.e. bilateral tubal ligation), iii) partner’s placement of an intrauterine device or intrauterine system, iv) partner’s use of hormonal contraception (implantable, patch, oral, vaginal ring), v) 2 barrier methods (their partner’s use of an occlusive cap [diaphragm, or cervical/vault caps] with spermicidal foam/gel/film/cream/suppository and the subject’s use of a condom) b) Male subjects who declare themselves as sexually abstinent are acceptable for the purposes of this study. Abstinence in this study is defined as “true abstinence: when this is in line with the preferred and usual lifestyle of the subject.” Any man making this declaration must consent to using the aforementioned highly effective contraceptive methods (as applicable) prior to engaging in sexual activity with female partners through 90 days after the last dose of study drug. c) All male subjects must abstain from sperm donation for the study period and for 90 days after the last dose of study drug. 7) Provide written informed consent. 8) In the opinion of the Investigator, the subject is able to adequately comply with all required study procedures for the duration of the study. The subject must demonstrate adequate compliance with all study procedures required including diary recording of HAE attacks.

1.Maschi o femmine non in stato di gravidanza o in allattamento, = 18 anni di età 2. Trattamento terapeutico con avoralstat completato precedentemente con successo OPPURE una diagnosi confermata da laboratorio di HAE di tipo 1 o 2. 3. Accordo da parte dello sperimentatore e il soggetto che il soggetto è un candidato valido per la profilassi da attacchi HAE. 4. Accesso appropriato a medicazioni per il trattamento di attacchi acuti di HAE. 5. Le partecipanti di sesso femminile devono presentare almeno uno dei seguenti requisiti: a. soggetto fertile (definito come donna in non-menopausa, che non ha subito una isterectomia, ovariectomia bilaterale, oppure manifesta un mal funzionamento documentato delle ovaie) che accetta di usare un metodo contraccettivo altamente efficace per l´intera durata dello studio e per 30 giorni dopo l´uso del farmaco. I seguenti metodi sono considerati metodi contraccettivi ad alta efficacia: • Sterilizzazione chirurgica (ie chiusura bilaterale delle tube) • Sterilizzazione chirurgica del partner (ie vasectomia) • Collocazione di Device intrauterino (IUD) oppure sistema intrauterino • Contraccettivo ormonale (impianto, cerotto, orale, anello vaginale) • Metodo a due barriere (il loro partner userà un profilattico e la partecipante un cappuccio (diaframma, oppure cappuccio cervicale) abbinato a spermicida a base di crema/schiuma/film/gel/supposta. Il soggetto in post-menopausa da = 2 anni e che ha un livello di FSH = 40 mIU/mL deve accettare di usare un metodo contraccettivo altamente efficace come accennato nel paragrafo superiore per l’intero periodo dello studio e per 30 giorni dopo l’intake dell’ultima dose. b. soggetto non fertile (definito come post-menopausa per > 2 anni oppure se in post-menopausa da = 2 anni e livello di FSH > 40mlI/mL , oppure una donna che ha avuto una isterectomia, ovariectomia bilaterale, oppure manifesta un malfunzionamento documentato delle ovaie). c. soggetto che dichiara di essere sessualmente astinente o che ha solo partner sessuali donne. L’astinenza in questo studio è definita come “vera astinenza: se questa è in linea con lo stile di vita usuale e preferita dal paziente”. Tutte le donne fertili che fanno questa dichiarazione devono acconsentire l’uso di un metodo contraccettivo altamente efficace prima di avere un qualsiasi contatto sessuale con un partner maschio per i 30 giorni dopo l’ intake dell’ultima dose. 6. I partecipanti di sesso maschile devono rispettare i seguenti requisiti: a. i soggetti maschi sessualmente attivi devono accettare l’utilizzo di un contraccettivo altamente efficace con la loro partner femminile se questa si trova in uno stato fertile (definita come post-menopausa da = 2 anni oppure se in non-menopausa che non ha avuto una isterectomia, ovariectomia bilaterale, oppure manifesta un malfunzionamento documentato delle ovaie) durante l’intero periodo dello studio e fino 90 giorni dopo l´ultimo dosaggio del farmaco studiato. I seguenti metodi sono considerati metodi contraccettivi ad alta efficacia: • Soggetto che ha subito una vasectomia • Sterilizzazione chirurgica della partner (ie chiusura bilaterale delle tube) • Partner che fa uso di un dispositivo oppure sistema intrauterino • Partner che fa uso di un contraccettivo ormonale (impianto, cerotto, orale, anello vaginale) • Metodo a due barriere (la loro partner farà uso di un cappuccio (diaframma, oppure cappuccio cervicale con spermicida a base di schiuma/gel/crema/film/supposta e il soggetto di un profilattico) b. i soggetti maschili che dichiarano di essere sessualmente astinente sono accettati per questo studio. L’astinenza in questo studio è definita come “vera astinenza: se questa è in linea con lo stile di vita usuale e preferita dal paziente”. Tutti i soggetti maschii che fanno questa dichiarazione devono acconsentire l’uso di uno dei metodi contraccettivi menzionati prima altamente efficaci prima di avere un qualsiasi contatto sessuale con un partner maschio per i 30 giorni dopo l’ intake dell’ultima dose. c. tutti i soggetti maschili devono astenersi dalla donazione di liquido seminale durante l’intero periodo dello studio e fino a 90 giorni dopo l’ ultimo dosaggio del farmaco studiato. 7. Firma del consenso informato scritto. 8. Secondo il parere dello sperimentatore, il soggetto é in grado di seguire tutte le procedure richieste durante l’ intera durata dello studio. Il soggetto deve dimostrare una adeguata complianza con tute le procedure richieste, inclusa la registrazione degli attacchi HAE nel diario.

E.4 | Principal exclusion criteria |
1) Females who are pregnant (positive urine or serum pregnancy test) or breast feeding at the screening visit or who are planning to become pregnant during the study period. 2) Any clinically significant medical condition or medical history that, in the opinion of the Investigator or Sponsor, would interfere with the subject’s safety or ability to participate in the study. 3) Any significant comorbid condition that is not controlled (e.g., gastrointestinal disease that leads to malabsorption) 4) Dementia, altered mental status or any psychiatric condition, or stay in an institution further to an official or court order that would prohibit the understanding or rendering of informed consent or participation in the study. 5) Current or past medical history of chronic pancreatitis, pancreatic insufficiency at any time in the past, or a confirmed history of acute pancreatitis occurring within the past 5 years. 6) Clinically significant abnormal ECG as assessed by the Investigator. 7) Any abnormal laboratory or urinalysis finding at the screening visit that, in the opinion of the Investigator or Sponsor, is clinically significant and relevant for this study. 8) Investigational drug exposure in any other investigational drug study within 30 days prior to the screening visit (with the exception of an investigational study of avoralstat). 9) History of alcohol or drug abuse within the previous year, or current evidence of substance dependence or abuse (self-reported alcoholic intake > 3 units alcohol/day). 10) For subjects undergoing a screening visit, positive drugs of abuse screen (unless as used as medical treatment, e.g., with a prescription). 11) For subjects undergoing a screening visit, infection with hepatitis B virus (HBV), hepatitis C virus (HCV), or human immunodeficiency virus (HIV). |
1.Donne in stato di gravidanza (test di gravidanza basato sull´urina oppure sul siero) oppure che allattano durante la visita di screening oppure che pianificano di rimanere gravide durante lo studio. 2. Qualsiasi condizione clinicamente significante o storia medica che, a parere dello sperimentatore oppure del promotore,potrebbe interferire con la sicurezza del paziente o la possibilitá di partecipare allo studio. 3. Qualsiasi condizione di cormobidità significativa non controllabile ( per esempio malattie gastrointestinali che portano a malassorbimento) 4. Demenza, disturbi mentali o qualsiasi condizioni psichiatrica, oppure la permanenza in un istituto ulteriormente ad un´ingunzione che impediscono la comprensione del consenso informato e la partecipazione allo studio. 5. Storia medica attuale o passata che riporta una pancreatite cronica, insufficienza pancreatica in passato, oppure la conferma di pancreatite acuta verificatasi negli ultimi 5 anni. 6. ECG clinicamente significativamente anormale valutato dallo sperimentatore. 7. Valori di laboratorio (siero e urina) definiti anormali e clinicamente significanti, secondo il parere dello sperimentatore oppure del promotore, durante la visita di screening 8. Attuale partecipazione a qualsiasi altro studio clinico oppure entro 30 giorni prima della visita di screening (con eccezione del farmaco oggetto dello studio avoralstat) 9. Storia clinica riguardo l´abuso di alcol oppure sostanze stupefacenti durante l´anno passato, oppure attuali segni di dipendenza o abuso da sostanze (dichiarando assunzione superiore a 3 unità di alcol al giorno) 10. Soggetti che risultano positivi durante la visita di screening alle sostanze stupefacenti (ad eccezione dei trattamenti medici, per esempio previsti di prescrizione medica) 11. Soggetti che presentano un infezione virale da epatite B ( HBV), epatite C (HCV), HIV durante la visita di screening.
