-To evaluate in children 6 to <12 years of age with uncontrolled persistent asthma: -The safety and tolerability of dupilumab. -The effect of dupilumab in improving patient-reported outcomes (PROs). -The dupilumab systemic exposure and incidence of anti-drug antibodies. -To explore the association between dupilumab treatment and pediatric immune responses to vaccines: any vaccination for tetanus, diphtheria, pertussis and/or seasonal trivalent/quadrivalent influenza vaccine. |

E.3 | Principal inclusion criteria |
-Children 6 to <12 years of age, with a physician diagnosis of persistent asthma for ≥12 months prior to Screening, based on clinical history and examination, pulmonary function parameters according to Global initiative for asthma (GINA) 2015 Guidelines and the following criteria: -Existing background therapy of medium-dose inhaled corticosteroids (ICS) with second controller medication (ie, long-acting β2 agonist [LABA], leukotriene receptor antagonist [LTRA], long acting muscarinic antagonist [LAMA], or methylxanthines) or high-dose ICS alone or high dose ICS with second controller, for at least 3 months with a stable dose ≥1 month prior to Screening Visit 1. -Pre-bronchodilator forced expiratory volume in 1 second (FEV1) ≤95% of predicted normal or pre bronchodilator FEV1/forced vital capacity (FVC) ratio <0.85 at Screening and Baseline Visits. -Reversibility of at least 10% in FEV1 after the administration of albuterol/salbutamol or levalbuterol/levosalbutamol reliever medication before randomization. Documented reversibility or positive airway hyperresponsiveness to methacholine within 12 months prior to Screening V1 is considered acceptable. -Must have experienced within 1 year prior to Screening Visit 1 a Severe Exacerbation Event. - Evidence of uncontrolled asthma, with at least one of the following criteria during the 4 (±1) weeks Screening Period: -Asthma Control Questionnaire–Interviewer Administered (ACQ-IA) ACQ-5 score ≥1.5 on at least one day of the Screening Period. -Use of reliever medication (ie, albuterol/salbutamol or levalbuterol/levosalbutamol), other than as a preventive for exercise induced bronchospasm, on 3 or more days per week, in at least one week during the Screening Period. -Sleep awakening due to asthma symptoms requiring use of reliever medication at least once during the Screening Period. -Asthma symptoms 3 or more days per week in at least one week during the Screening Period. |
Niños de 6 a <12 años de edad, con diagnóstico por parte del Investigador de asma persistente durante ≥12 meses antes de la selección, basado en el examen y los antecedentes clínicos, los parámetros de función pulmonar según las directrices de la Iniciativa global para el asma (GINA) de 2015 y los siguientes criterios: - Tratamiento de base existente con una dosis media de ICS más un segundo medicamento de control (es decir, LABA, LTRA, LAMA o metilxantinas) o una dosis alta de ICS en monoterapia o una dosis alta de ICS más un segundo medicamento de control, durante al menos 3 meses con una dosis estable ≥1 mes antes de la Visita de Selección 1. - Volumen espiratorio forzado en un segundo (VEF1) prebroncodilatador ≤95 % del valor normal previsto o proporción VEF1 prebroncodilatador/capacidad vital forzada (CVF) <0,85 en las Visitas de Selección y Basal. - Reversibilidad de al menos un 10 % en el VEF1 después de la administración de 200 mcg a 400 mcg (2 a 4 inhalaciones con inhalador de dosis medida [IDM]) de albuterol/salbutamol o 45 mcg a 90 mcg (2 a 4 inhalaciones con IDM) levalbuterol/levosalbutamol como medicación de alivio antes de la aleatorización (se permiten hasta 3 oportunidades durante la misma visita y un máximo de 12 inhalaciones de medicación de alivio si el paciente las tolera). Se considera aceptable la documentación de reversibilidad o hiperreactividad positiva de las vías respiratorias a la metacolina en los 12 meses anteriores a la V1 de Selección. - Haber experimentado, en el año anterior a la Visita 1 de Selección, alguno de los siguientes eventos: a) Tratamiento con un corticoesteroide sistémico (CES, oral o parenteral), según lo prescrito por un profesional sanitario para tratar el empeoramiento del asma al menos una vez, u b) Hospitalización o visita al servicio de urgencias por empeoramiento del asma. - Evidencias de asma no controlada, con al menos uno de los criterios siguientes durante el período de selección de 4 (±1) semanas: a) Puntuación del Cuestionario de Control del Asma Administrado por el Entrevistador (ACQ-IA) ACQ-5 (Cuestionario de Control del Asma Administrado por el Entrevistador, versión de 5 preguntas) ≥1,5 al menos durante un día del período de selección. b) Uso de medicación de alivio (es decir, albuterol/salbutamol o levalbuterol/levosalbutamol), distinto del preventivo para el broncoespasmo de esfuerzo, 3 días a la semana o más, al menos durante una semana del Período de Selección. c) Despertar durante el sueño a causa de los síntomas del asma que requiere el uso de medicación de alivio al menos una vez durante el Período de Selección. d) Síntomas de asma 3 días a la semana o más al menos durante una semana del Período de Selección. |
