E.3 | Principal inclusion criteria |
The patients eligible for inclusion in this study must fulfill all of the following criteria:
1. Written informed consent must be obtained before any assessment is performed.
2. Male and female subjects aged 18 to 45 years of age inclusive, and otherwise in good health as determined by medical history, physical examination, and vital signs.
3. Body weight between 50 and 120 kg, both inclusive, at screening.
4. Patients with papulo-pustular acne vulgaris (inflammatory acne):
- presenting at baseline with :
• 20 to 100 facial inflammatory lesions (papules, pustules and nodules),
• presenting at baseline and screening with
• no more than 2 facial inflammatory nodules or cysts,
• and a minimum number of 10 non-inflammatory facial lesions (open and closed comedones)
- who are candidates for systemic treatment and for whom in the opinion of the investigator, an appropriate previous treatment with topical anti-acne medication :
• failed,
• or was not well tolerated,
• or is not indicated (e.g., due to large body surface area affected, e.g., on the back)
5. Patients with Grade 3 (moderate) or Grade 4 (severe) IGA score confirmed by central reading of standardized image capture (Visia® system) by an independent dermatologist at screening and by the investigator’s clinical evaluation at baseline.
6. Able to communicate well with the investigator, to understand and comply with the requirements of the study.

Les patients éligibles pour inclusion dans cette étude doivent réunir tous les critères suivants :
1. Obtention du consentement éclairé écrit avant toute évaluation.
2. Hommes et femmes âgés de 18 à 45 ans révolus, et par ailleurs en bonne santé selon les antécédents médicaux, l'examen clinique et les signes vitaux.
3. Poids corporel compris entre 50 kg et 120 kg à la sélection.
4. Patients atteints d’acné vulgaris papulo-pustuleuse (acné inflammatoire) :
• présentant à la baseline :
• 20 à 100 lésions inflammatoires faciales (papules, pustules et nodules),
• présentant à la baseline et à la sélection :
• pas plus de 2 nodules ou kystes inflammatoires faciaux,
• et un nombre minimum de 10 lésions faciales non inflammatoires (comédons ouverts et fermés),
• qui sont candidats à un traitement systémique et pour qui, de l'avis de l'investigateur, un traitement préalable approprié par un médicament anti-acnéique topique :
• a échoué,
• ou n'a pas été bien toléré,
• ou n'est pas indiqué (par ex. en raison de la taille de la surface corporelle affectée, par ex. sur le dos).
5. Patients dont le score sur l’échelle IGA est de grade 3 (modéré) ou de grade 4 (sévère) confirmé par une interprétation centralisée de la capture d'image standardisée (système Visia®) réalisée par un dermatologue indépendant à la sélection et par l'évaluation clinique de l'investigateur à la baseline).
6. Etre en mesure de bien communiquer avec l'investigateur, de comprendre et respecter les exigences de l'étude. |

E.4 | Principal exclusion criteria |
The patients fulfilling any of the following criteria are not eligible for inclusion in this study:
1. Previous treatment with investigational drugs at the time of screening, or within 4 weeks or 5 half-lives of baseline, whichever is longer; or more as required by local regulations.
2. Previous treatment with any topical anti-acne therapy:
• prescription treatment within 2 weeks prior to baseline
• OTC within 1 week prior to baseline
The use of medicated anti-acne creams, medicated cleansers or medicated soaps is prohibited.
3. Previous treatment with any oral/systemic anti-acne therapy:
• oral antibiotics, dapsone, oral zinc within 4 weeks prior to baseline,
•retinoids, within 3 months prior to baseline and
• hormonal therapy (within 1 month prior to baseline.
If women of child bearing potential are using oral contraception, this contraception can be used under certain conditions.
4. Previous treatment with systemic corticosteroids or immunomodulators (e.g. cyclosporine, methotrexate, azathioprine) within 4 weeks prior to baseline.
5. Previous treatment with biologics (e.g anti-TNFα agents, anti-IL-1, or anti-IL-17) within 3 months or 5 half-lives (whichever is longer) prior to baseline.
6. Previous treatment with anti-IL-12/23 blocking agents (e.g. briakinumab and ustekinumab or p19 antibodies) within 6 months prior to baseline.
7. Previous surgical, physical (such as ThermaClear™), light (including blue or UV light, photodynamic therapy) or laser therapy within 4 weeks prior to baseline.
8. Previous facial treatment with medium depth chemical peels (excluding home regimens) within 3 months prior to baseline.
9. Concomitant medication(s) known to inhibit OAT3 or BCRP and that cannot be discontinued or replaced by safe alternative medication within 5 half-lives or 1 week (whichever is longer) to baseline and for the duration of the study.
10. Any other forms of acne.
11. Any severe, progressive or uncontrolled medical or psychiatric condition or other factors at randomization that in the judgment of the investigator prevents the patient from participating in the study.
12. Active systemic infections (other than common cold) within 2 weeks prior to baseline.
13. Subjects with eGFR <60 mL/min/1.73m2 at screening.
14. History or presence of crystals or stones in urine.
15. History or symptoms of malignancy of any organ system, treated or untreated, within the past 5 years.
16. Chronic infection with Hepatitis B or C.
17. History of auto-immune or immunodeficiency diseases, or a positive HIV test result at screening.
18. Pregnant or nursing women, where pregnancy is defined as the state of a female after conception and until the termination of gestation, confirmed by a positive hCG laboratory test.
19. Women of child-bearing potential, unless they are using basic methods of contraception during dosing of study treatment.
20. History of drug abuse or unhealthy alcohol use.
21. Donation or loss of 400 ml or more of blood within 8 weeks prior to baseline, or longer if required by local regulation.
22. Inability or unwillingness to undergo repeated venipunctures.
23. Any surgical or medical condition which might significantly alter the absorption, distribution, metabolism, or excretion of drugs, or which may jeopardize the subject in case of participation in the study.
Other protocol-defined exclusion criteria may apply.

Les patients remplissant l’un des critères suivants ne sont pas éligibles pour inclusion dans cette étude :
1. Traitement antérieur par d’autres médicaments à l'étude au moment de la sélection ou dans les 4 semaines ou les 5 demi-vies du médicament précédant la sélection, la durée la plus longue prévalant, ou un délai plus long si les réglementations locales l’exigent.
2. Traitement antérieur par tout traitement anti-acnéique topique :
• traitement sur ordonnance dans les 2 semaines précédant la baseline,
• traitement en vente libre (OTC) dans la semaine précédant la baseline.
L'utilisation de crèmes anti-acnéiques médicamenteuses, de nettoyants médicamenteux ou de savons médicamenteux restera interdite pendant la durée de l'étude.
3. Traitement antérieur par tout traitement anti-acnéique oral/systémique :
• antibiotiques oraux, dapsone, zinc par voie orale dans les 4 semaines précédant la baseline,
• rétinoïdes, par ex. isotrétinoïne, dans les 3 mois précédant la baseline,
• hormonothérapie dans le mois précédant la baseline.
Si les femmes en âge de procréer utilisent une contraception orale, cette contraception peut être utilisée sous certaines conditions.
4. Traitement antérieur par corticoïdes systémiques ou immunomodulateurs (tels que, mais sans s'y limiter, cyclosporine, méthotrexate, azathioprine) dans les 4 semaines précédant la baseline.
5. Traitement antérieur par des agents biologiques (tels que les agents anti-TNFα, anti-IL-1 ou anti-IL-17) dans les 3 mois ou 5 demi-vies (la durée la plus longue prévalant) précédant la baseline.
6. Traitement antérieur par des agents bloquants anti-IL-12/23 (tels que briakinumab et ustékinumab ou anticorps p19) dans les 6 mois précédant la baseline.
7. Traitement antérieur chirurgical, physique (tel que ThermaClear™), photothérapie (y compris exposition à la lumière bleue ou aux rayons UV, thérapie photodynamique [PDT]) ou traitement au laser dans les 4 semaines précédant la baseline.
8. Traitement facial antérieur par des exfoliations chimiques de profondeur moyenne (à l'exclusion des gommages faits maison) dans les 3 mois précédant la baseline.
9. Patients recevant un ou des médicaments concomitants connus pour inhiber l'OAT3 ou la BCRP (voir le Tableau 5-1 du protocole) et qui ne peuvent pas être interrompus ou remplacés par des médicaments alternatifs sûrs dans les 5 demi-vies du médicament ou 1 semaine (la durée la plus longue prévalant) précédant la baseline et pendant toute la durée de l'étude.
10. Toute autre forme d'acné
11. Toute affection médicale ou psychiatrique sévère, progressive ou incontrôlée ou tout autre facteur lors de la randomisation qui, de l’avis de l'investigateur, empêche le patient de participer à l'étude.
12. Antécédents d'hypersensibilité ou d'allergie au composé à l'étude et aux inhibiteurs de LTA4H ou LTB4 utilisés dans cette étude.
13. Patients présentant un débit de filtration glomérulaire estimé (DFGe) <60 mL/min/1,73 m2 à la sélection.
14. Antécédents ou présence de cristaux ou de calculs dans l'urine
15. Antécédents ou symptômes de tumeur maligne affectant tout système d’organes, traitée ou non, au cours des 5 dernières années
16.Infection chronique par le virus de l'hépatite B (VHB) ou de l'hépatite C (VHC)
17. Antécédents de maladies auto-immunes ou d'immunodéficience, ou résultat positif au test du virus de l'immunodéficience humaine (VIH) (ELISA et Western blot) à la sélection.
18. Grossesse ou allaitement, la grossesse étant définie comme l'état de la femme après la conception et jusqu'au terme de la gestation, confirmé par un dosage biologique positif d'hCG.
19 Femmes en âge de procréer, c'est-à-dire les femmes physiologiquement aptes à être enceinte, sauf si elles utilisent des moyens de contraception efficaces pendant l'administration du traitement à l'étude.
20. Antécédents de toxicomanie ou d’alcoolisme
21. Don de sang ou perte de sang d’au moins 400 ml dans les 8 semaines précédant la baseline, ou dans un délai plus long si les législations locales l’exigent.
22. Incapacité ou refus de subir des ponctions veineuses répétées (par ex. en raison d’un problème de tolérance ou d’abord veineux).
23. Toute affection chirurgicale ou médicale susceptible de modifier significativement l’absorption, la distribution, le métabolisme ou l’excrétion des médicaments, ou pouvant mettre le patient en danger en cas de participation à l’étude. |
