    The EU Clinical Trials Register currently displays   43865   clinical trials with a EudraCT protocol, of which   7286   are clinical trials conducted with subjects less than 18 years old.   The register also displays information on   18700   older paediatric trials (in scope of Article 45 of the Paediatric Regulation (EC) No 1901/2006).

    To demonstrate the efficacy in subjects with T-PLL treated with venetoclax plus ibrutinib

    - To assess safety and tolerability in subjects with T-PLL treated with venetoclax plus ibrutinib,
    - To assess the ability of the subset of transplant-naïve subjects to proceed to further autologous/allogeneic stem cell transplantation (assessed by number of eligible subjects reaching transplant),
    - Valutare la sicurezza e la tollerabilità in soggetti affetti da T-PLL trattati con venetoclax in associazione a ibrutinib.
    - Valutare la capacità del sottogruppo di soggetti naïve al trapianto di procedere a ulteriore trapianto autologo/allogenico di cellule staminali (valutato in base al numero di soggetti idonei che arrivano al trapianto).

    E.3Principal inclusion criteria
    - Male or female subjects, at least 18 years old, with a diagnosis of R/R T-PLL that requires treatment and suitable for oral administration of
    study drugs.
    - Subjects should meet the following disease activity criteria: an Eastern Cooperative Oncology Group performance status = 2.
    - Subjects should have laboratory values meeting the following criteria:
    • Alanine aminotransferase/aspartate aminotransferase = 3 × the upper limit of normal (ULN);
    • Adequate liver function as indicated by a total bilirubin = 1.5 x ULN (subjects with documented Gilbert's syndrome may have bilirubin > 1.5
    × ULN)
    • Absolute neutrophil count > 1000/µL;
    • Platelet count > 50,000/µL;
    • Creatinine clearance = 30 mL/minute; and
    • Hemoglobin > 8 g/dL.
    Soggetti di ambo i sessi, di età pari o superiore a 18 anni, con diagnosi di T-PLL R/R per cui sia necessario il trattamento e che siano idonei a ricevere i medicinali sperimentali per via orale.
    I soggetti devono soddisfare i seguenti criteri relativi all’attività della malattia: punteggio Eastern Cooperative Oncology Group relativo allo stato funzionale = 2.
    I soggetti devono presentare valori di laboratorio che soddisfano i seguenti criteri:
    • alanina aminotransferasi/aspartato aminotransferasi = 3 × il limite superiore della norma (ULN);
    • adeguata funzione epatica, confermata da valori di bilirubina totale = 1,5 x ULN

    • conta delle piastrine > 50.000/µL;

    Subjects may continue receiving venetoclax for upto 2 years provided
    they continue to tolerate the drug,have no evidence of disease
    progression (based on investigator assessment),do not have
    unacceptable toxicity,death,do not meet any of the criteria for subject
    discontinuation. After treatment discontinuation,subjects will be
    followed for disease progression (if not already occurred) and
    survival.Post-treatment calls will be performed every month until
    discontinuation from the study.
    I soggetti potranno continuare a ricevere venetoclax per massimo 2 anni se continuano a tollerarlo,in assenza di evidenza di progressione (in base alla valutazione del PI),di tossicità,decesso,se non presentano alcuno dei criteri che richiedono l’interruzione.Una volta interrotto il trattamento,i soggetti saranno seguiti per rilevare la progressione(se non già avvenuta)e la sopravvivenza. I contatti telefonici post trattamento avranno luogo ogni mese fino all’interruzione della partecipazione
