E.3 | Principal inclusion criteria |
1. Subject is = 18 years of age at the time of signing the informed consent form (ICF). 2. Subject with histological or cytological confirmation of extensive stage SCLC (ES SCLC) or Stage IIIb or IV sqNSCLC. 3. Subject has received 1 or 2 prior lines of therapie (further defined in the protocol). 4. Subject has progressed at the last line of therapy. 5. Subject has a measurable disease defined by RECISTv1.1. 6. Subject agrees to provide a tumor biopsy from primary or metastatic site prior to first dose and at a pre-specified timepoint during treatment. -Refer to the protocol for highly effective contraceptive methods. c. Avoid conceiving or donating ova while on treatment and for 45 days after the last dose of CC-90011 or 5 months after the last dose of nivolumab, whichever is later. d. Agree to ongoing pregnancy testing during the course of the study. This applies even if the subject practices true abstinence* from heterosexual contact. Core biopsy is required however, in the event a core biopsy may not otherwise be feasible in the opinion of the treating physician, an endobronchial ultrasound-guided fine needle aspirate [EBUS-FNA]) biopsy, using the largest gauge needle, may be performed instead. 7. Subject has ECOG Performance Status of 0 to 1. 8. Subject is able to swallow medication. 9. Subject must have the lab values as specified in the protocol. 10.A female of childbearing potential (FCBP) is a female who: - has achieved menarche at some point, - has not undergone a hysterectomy or bilateral oophorectomy, or - has not been naturally postmenopausal (amenorrhea following cancer therapy or other medical condition does not rule out childbearing potential) for at least 24 consecutive months (ie, has had menses at any time in the preceding 24 consecutive months) and must: a. Have 2 negative pregnancy tests as verified by the Investigator prior to starting study treatments as specified in the protocol. b. Either commit to true abstinence from heterosexual contact (which must be reviewed on a monthly basis and source documented) or agree to use, and be able to comply with 1 highly effective contraceptive method plus 1 barrier method during the following time periods related to this study: 1) from signing of ICF; 2) while taking study treatment; 3) during dose interruptions; and 4) for at least 45 days after the subject's last dose of CC-90011 or 5 months after the last dose of nivolumab, whichever is later. -Refer to the protocol for highly effective contraceptive methods. c. Avoid conceiving or donating ova while on treatment and for 45 days after the last dose of CC-90011 or 5 months after the last dose of nivolumab, whichever is later. d. Agree to ongoing pregnancy testing during the course of the study. This applies even if the subject practices true abstinence* from heterosexual contact. 11.Males must practice true abstinence from heterosexual intercourse (which must be reviewed on a monthly basis) or agree to use a condom (a latex or non-latex synthetic condom is recommended)during sexual contact with a pregnant female or a FCBP while participating in the study, during dose interruptions, and for at least 105 days after the subject's last dose of CC-90011 or 7 months after the last dose of nivolumab, whichever is later, even if he has undergone a successful vasectomy. Males must agree not to donate semen or sperm while on treatment and for at least 105 days following the last dose of CC-90011 or 7 months after the last dose of nivolumab, whichever is later. 12.Subject must understand and voluntarily sign an ICF prior to any study-related assessments/procedures being conducted. 13.Subject is willing and able to adhere to the study visit schedule and other protocol requirements. |

Il soggetto acconsente a fornire una biopsia tumorale dalla sede primaria o metastatica prima della prima dose e in punti temporali prestabiliti durante il trattamento. È richiesta una biopsia percutanea; tuttavia, nel caso in cui la biopsia percutanea non sia fattibile a parere del medico curante, può essere eseguita invece un’ecografia endobronchiale con agoaspirato [EBUS-FNA], usando l’ago con il calibro maggiore. 7. Il soggetto ha un performance status ECOG (Eastern Cooperative Oncology Group) tra 0 e 1. 8. Il soggetto è in grado di deglutire il medicinale. 9. Il soggetto deve avere i valori di laboratorio specificati nel protocollo. 10. Un donna potenzialmente fertile (FCBP) è una donna che: - ha già avuto le mestruazioni, - non è stata sottoposta ad isterectomia o ovariectomia bilaterale, o - non è in menopausa naturale (l’amenorrea in seguito alla terapia antitumorale o altra condizione medica non esclude la fertilità) da almeno 24 mesi consecutivi (cioè ha avuto le mestruazioni in qualsiasi momento nei precedenti 24 mesi consecutivi) e deve: a. Presentare 2 test di gravidanza negativi verificati dallo Sperimentatore prima di iniziare i trattamenti in studio, come specificato nel protocollo. b. Impegnarsi all’astinenza completa da contatti eterosessuali (che deve essere rivalutata mensilmente e documentata) o acconsentire ad usare in modo costante 1 metodo contraccettivo altamente efficace più 1 metodo a barriera durante i seguenti periodi relativi allo studio: 1) dalla firma dell’ICF; 2) mentre assume il trattamento in studio; 3) durante le interruzioni di dose; e 4) per almeno 45 giorni dopo l’ultima dose di CC-90011 o 5 mesi dopo l’ultima dose di nivolumab, in base all’evento che si verifica più tardi. -Consultare il protocollo per i metodi contraccettivi altamente efficaci. c. Evitare di concepire o donare ovuli durante il trattamento e per 45 giorni dopo l’ultima dose di CC-90011 o 5 mesi dopo l’ultima dose di nivolumab, in base all’evento che si verifica più tardi. d. I soggetti di sesso maschile devono praticare astinenza completa da rapporti eterosessuali (che deve essere rivalutata mensilmente) o acconsentire ad usare un preservativo (si raccomanda un preservativo sintetico di lattice o non di lattice) durante il contatto sessuale con una donna in stato di gravidanza o con una donna potenzialmente fertile durante la partecipazione allo studio, durante le interruzioni di dose e per almeno 105 giorni dopo l’ultima dose di CC-90011 o 7 mesi dopo l’ultima dose di nivolumab, in base all’evento che si verifica più tardi, anche se il soggetto fosse stato sottoposto ad un intervento riuscito di vasectomia. I soggetti di sesso maschile devono acconsentire a non donare seme o sperma durante il trattamento e per 105 giorni dopo l’ultima dose di CC-90011 o 7 mesi dopo l’ultima dose di nivolumab, in base all’evento che si verifica più tardi. 12. Il soggetto deve capire e firmare volontariamente un ICF prima che siano condotte valutazioni/procedure connesse allo studio. 13. Il soggetto è disposto e in grado di aderire al piano di visite dello studio e ad altri requisiti del protocollo. |

1. Subject has any significant medical condition, laboratory abnormality, or psychiatric illness that would prevent the subject from participating in the study. 2. Subject has any condition including the presence of laboratory abnormalities, which places the subject at unacceptable risk if he/she were to participate in the study. 3. Subject has any condition that confounds the ability to interpret data from the study. 4. Subject has not recovered to Grade 2 or lower clinically significant toxicities related to the prior therapy (alopecia excluded). 5. Subject has received prior LSD1 therapies. 6. Subject has a history of severe hypersensitivity reactions to other monoclonal antibodies. 7. Subject with symptomatic and untreated or unstable central nervous system (CNS) metastases. 8. Subject has persistent diarrhea due to a malabsorptive syndrome (such as celiac sprue or inflammatory bowel disease) = NCI CTCAE Grade 2, despite medical management), or any other significant gastrointestinal (GI) disorder that could affect the absorption of the study treatments. 9. Subject with symptomatic or uncontrolled ulcers (gastric or duodenal), particularly those with a history of and/or risk of perforation and GI tract hemorrhages. 10.Subject with any hemorrhage/bleeding event > NCI CTCAE Grade 2 or haemoptysis > 1 teaspoon within 4 weeks prior to the first dose. 11.Subject has any of the following cardiovascular criteria further specified in the protocol. 12.Subject has known human immunodeficiency virus (HIV) infection. 13.Subject has known chronic active hepatitis B or C virus (HBV, HCV) infection. a. Subject who is seropositive due to HBV vaccination is eligible. b. Subject who has no active viral infection and is under adequate prophylaxis against HBV reactivation is eligible. 14.Subject has any other malignancy within 2 years prior to enrollment, with the exception of adequately treated in-situ bladder cancer, in-situ carcinoma of the cervix, uteri, nonmelanomatous skin cancer, ductal in situ breast carcinoma, thyroid cancer, or early stage prostate cancer (all treatment of which should have been completed 6 months prior to enrollment). 15.Enrollment in any other clinical protocol or investigational study with an interventional agent or assessments that may interfere with study procedures. 16.Subject has medical conditions requiring systemic treatment with either corticosteroids (> 10 mg daily prednisone or equivalent) or other immunosuppressive medications within 14 days of enrollment. a. A brief (= 7 days) course of corticosteroids for prophylaxis (eg, contrast dye allergy) or for treatment of nonautoimmune conditions (eg, delayed-type hypersensitivity reaction caused by contact allergen) is permitted. b. Adrenal replacement steroid doses > 10 mg daily prednisone or equivalent are permitted in the absence of active autoimmune disease. c. Topical, ocular, intra-articular, intranasal, and inhalational corticosteroids (with minimal systemic absorption) are permitted. 17.Subject has active autoimmune diseases or history of autoimmune diseases that may relapse. Subjects with the following diseases are allowed to be enrolled after further screening: type I diabetes, hypothyroidism managed with hormone replacement therapy only, skin diseases not requiring systemic treatment (such as vitiligo, psoriasis, or alopecia), or diseases not expected to recur in the absence of external triggering factors. 18.Subject is pregnant or nursing. ... Il s. presenta qualsiasi condizione, compresa la presenza di anomalie dei test di laboratorio, che lo/la esporrebbe a un rischio inaccettabile nel caso in cui partecipasse allo studio. 3. Il s. presenta qualsiasi condizione che confonda la capacità di interpretare i dati provenienti dallo studio. 4. Il s. non è guarito da tossicità clinicamente significative di Grado 2 o inferiore relative alla terapia precedente (alopecia esclusa). 5. Il s. ha ricevuto precedenti terapie LSD1 6. Il s. ha un’anamnesi di gravi reazioni di ipersensibilità ad altri anticorpi monoclonali. 7. S.con metastasi sintomatiche e non trattate o instabili del sistema nervoso centrale (SNC). 8. Il s. ha diarrea persistente dovuta a sindrome di malassorbimento (come sprue celiaco o malattia infiammatoria intestinale) = Grado 2 NCI CTCAE, nonostante il trattamento medico, o ha qualsiasi altro disturbo gastrointestinale (GI) significativo che potrebbe influenzare l’assorbimento dei trattamenti in studio. 9. S. con ulcere sintomatiche o non controllate (gastriche o duodenali), in particolare i soggetti con anamnesi e/o rischio di perforazione ed emorragie del tratto GI. 10. S. con qualsiasi evento di emorragia/sanguinamento > Grado 2 NCI CTCAE o emoptisi > 1 cucchiaino entro 4 settimane precedenti la prima dose. 11. Il s.presenta uno qualunque dei seguenti criteri cardiovascolari ulteriormente specificati nel protocollo. 12. Il s.ha un’infezione nota del virus dell’immunodeficienza umana (HIV). 13. Il soggetto ha un’infezione attiva cronica nota da virus dell’epatite B o C (HBV, HCV). a. Il s. che è sieropositivo a causa della vaccinazione per HBV è idoneo. b. Il s.che non ha alcuna infezione virale attiva ed è sotto profilassi adeguata contro la riattivazione di HBV, è idoneo. 14. Il s.ha avuto qualsiasi altra neoplasia maligna entro 2 anni prima dell’arruolamento, con l’eccezione del cancro della vescica adeguatamente trattato in-situ, carcinoma in-situ delle cervice, dell’utero, carcinoma cutaneo non melanomatoso, carcinoma mammario duttale in-situ, cancro della tiroide, o cancro della prostata in stadio precoce (per il quale tutto il trattamento deve essere stato completato 6 mesi prima dell’arruolamento). 15. Arruolamento in qualsiasi altro protocollo clinico o studio sperimentale con un agente o valutazione interventistico che potrebbe interferire con le procedure dello studio. 16. Il soggetto ha una condizione medica che richieda trattamento sistemico con corticosteroidi (> 10 mg al giorno di prednisone o equivalente) o altri medicinali immunosoppressori entro 14 giorni dall’arruolamento. a. È consentito un breve (= 7 giorni) corso di corticosteroidi per profilassi (ad es. allergia al mezzo di contrasto) o per il trattamento di malattie non autoimmuni (ad es. reazioni di ipersensibilità di tipo ritardato provocate da contatto con allergene). b. Dosi di steroidi per la terapia sostitutiva adrenergica > 10 mg al giorno di prednisone o equivalente sono concessi in assenza di malattia autoimmune attiva. c.
