E.3 | Principal inclusion criteria |
- Subject: male or female aged ≥ 18 years and ≤ 75 years - Diagnosis of non-segmental (symmetrical) vitiligo with a body surface area involved >5% excluding hands and feet - Active non-segmental vitiligo is defined by: • Non-segmental vitiligo with new patches or extension of old lesions during the last 6 months AND • Presence of hypochromic aspect under Wood’s lamp examination and/or perifollicular hypopigmentation under Wood’s lamp examination. - Able to read, understand, and give documented (electronic or paper signature) informed consent - Registered in the French Social Security - Agree to discontinue the use of the following excluded medications/treatments for at least 4 weeks prior to randomization (Visit 2) and throughout the study: systemic steroids, phototherapy, methotrexate, cyclosporine, mycophenolate mofetil, and azathioprine. - Agree to discontinue the use of the following excluded medications for at least 2 weeks prior to randomization (Visit 2) and throughout the study: • TCS or topical immune modulators (e.g., tacrolimus or pimecrolimus) • Topical phosphodiesterase type 4 (PDE-4) inhibitor (crisaborole) • Topical JAK inhibitor (e.g., tofacitinib or ruxolitinib) and/or any other investigational topical treatments.
- Patient characteristics - Are male or nonpregnant, nonbreastfeeding female patients, except: a. Male patients must agree to use 2 forms of birth control (1 must be highly effective, see below) while engaging in sexual intercourse with female partners of childbearing potential while enrolled in the study and for at least 4 weeks following the last dose of investigational product.
b. Female patients of childbearing potential must agree to use 2 forms of birth control, when engaging in sexual intercourse with a male partner while enrolled in the study and for at least 4 weeks following the last dose of investigational product. The following birth control methods are considered acceptable (the patient should choose 2 to be used with their male partner, and 1 must be highly effective): • Highly effective birth control methods: oral, injectable, or implanted hormonal contraceptives (combined estrogen/progesterone or progesterone only, associated with inhibition of ovulation); intrauterine device or intrauterine system (e.g., progestin-releasing coil); or vasectomized male (with appropriate post vasectomy documentation of the absence of sperm in the ejaculate). • Effective birth control methods: condom with a spermicidal foam, gel, film, cream, or suppository; occlusive cap (diaphragm or cervical/vault caps) with a spermicidal foam, gel, film, cream, or suppository; or oral hormonal contraceptives.
c. Females of nonchildbearing potential are not required to use birth control and they are defined as: • Women ≥60 years of age or women who are congenitally sterile, or • Women ≥40 and <60 years of age who have had a cessation of menses for ≥12 months and a follicle-stimulating hormone (FSH) test confirming nonchildbearing potential (≥40 mIU/mL or ≥40 IU/L), or women who are surgically sterile (i.e., have had a hysterectomy or bilateral oophorectomy or tubal ligation). - Signed informed consent form (ICF)

- Homme ou femme âgé de ≥ 18 ans et ≤ 75 ans - Diagnostic de vitiligo non segmentaire (symétrique) avec une surface corporelle impliquée >5% à l'exclusion des mains et des pieds - Le vitiligo actif non segmentaire est défini par : • Vitiligo non segmentaire avec nouvelles taches ou extension d'anciennes lésions au cours des 6 derniers mois ET • Présence d'un aspect hypochrome sous l'examen de la lampe de Wood et/ou d'une hypopigmentation périfolliculaire sous l'examen de la lampe de Wood. - Capacité à lire, comprendre et donner un consentement éclairé documenté (signature électronique ou sur papier) - Affilié ou bénéficiaire d’un régime de sécurité sociale - Accepter d’arrêter l'utilisation des médicaments/traitements exclus suivants pendant au moins les 4 semaines précédant la randomisation (visite 2) et pendant toute la durée de l'étude: corticothérapie générale, photothérapie, méthotrexate, cyclosporine, mycophénolate mofétil, azathioprine. - Accepter d’arrêter l'utilisation des médicaments exclus suivants pendant au moins les 2 semaines précédant la randomisation (visite 2) et pendant toute la durée de l'étude : - TCS ou modulateurs immunitaires topiques (par exemple, tacrolimus ou pimecrolimus) - Inhibiteur topique de la phosphodiestérase de type 4 (PDE-4) (crisaborole) - Inhibiteur topique de la voie JAK (par exemple, tofacitinib ou ruxolitinib) et/ou tout autre traitement topique expérimental.
Caractéristiques du patient - Hommes ou femmes non enceintes et n'allaitant pas: a. Les patients masculins doivent accepter d'utiliser 2 formes de contraception (1 doit être hautement efficace, voir ci-dessous) lors de leurs rapports sexuels avec des partenaires féminines en capacité de procréer pendant qu'ils participent à l'étude et pendant au moins 4 semaines après la dernière administration du produit de la recherche. b. Les patientes en capacité de procréer doivent accepter d'utiliser deux méthodes de contraception lorsqu'elles ont des rapports sexuels avec un partenaire masculin pendant qu'elles participent à l'étude et pendant au moins quatre semaines après la dernière administration du produit de la recherche. Les méthodes de contraception suivantes sont considérées comme acceptables (le patient doit en choisir 2 dont une hautement efficace) : - Méthodes de contraception hautement efficaces : contraceptifs hormonaux oraux, injectables ou implantés (combinaison oestrogène/progestérone ou progestérone seule, associée à une inhibition de l'ovulation) ; dispositif ou système intra-utérin (par exemple, stérilet libérant de la progestérone) ; ou vasectomie chez l’homme (avec documentation appropriée post-vasectomie de l'absence de spermatozoïdes dans l'éjaculat). - Méthodes efficaces de contraception : préservatif avec mousse, gel, film, crème spermicide ou suppositoire ; capuchon occlusif (diaphragme ou cape cervicale) avec mousse, gel, film, crème spermicide ou suppositoire ; ou contraceptifs hormonaux par voie orale.
c. - Segmented or mixed vitiligo - Patients who currently have or have a history of other concomitant skin conditions (e.g., psoriasis or lupus erythematosus) that could interfere with evaluations of the effect of the study drug on vitiligo - Patients who currently have a skin infection requiring treatment or who are currently being treated with topical or systemic antibiotics. Note: Patients can only be rescreened at least 4 weeks after the date of their previous screening failure and at least 2 weeks after resolution of the infection. - Patients who have severe concomitant disease that is expected to require the use of systemic steroids or otherwise interfere with study participation or require frequent active surveillance. (e.g., chronic unstable asthma). - Patients who have been treated with the following therapies: a) monoclonal antibodies (e.g. ustekinumab, omalizumab, dupilumab) for less than 5 half-lives prior to randomization. b) prior treatment with an oral JAK inhibitor (e.g., tofacitinib, ruxolitinib) c) systemic corticosteroid administered within 4 weeks prior to planned randomization or are expected to require systemic corticosteroids during the study. d) intra-articular corticosteroid injection within 4 weeks prior to the planned randomization date. e) more than 250 sessions of UV therapy. - Patients who are totally or partially unable to provide their own care (e.g., bed rest), such as being bedridden. - Patients who have uncontrolled hypertension characterized by repeated systolic blood pressure > 160 mm Hg or diastolic blood pressure > 100 mm Hg while seated. - Patients who have undergone major surgery within 8 weeks prior to screening or who will require major surgery during the course of the study that, in the opinion of the investigator, would present an unacceptable risk to the patient. - Patients who are immunocompromised and, in the opinion of the investigator, would present an unacceptable risk of participating in the study. - Patients who have had one of the following events within 12 weeks prior to the screening visit: venous thromboembolic event (VTE), myocardial infarction (MI), unstable ischemic heart disease, stroke, or NYHA Stage III/IV heart failure. - Patients with a history of recurrent VTE (≥ 2) or who are considered at high risk of VTE by the investigator. - Patients with a history of or presenting with cardiovascular, respiratory, hepatic, gastrointestinal, endocrine, hematological, neurological, or neuropsychiatric disorders or any other serious and/or unstable disease that, in the opinion of the investigator, could pose an unacceptable risk with the investigational product or interfere with the interpretation of the data. - Patients who have a history of lymphoproliferative disease; or have signs or symptoms suggestive of possible lymphoproliferative disease, including lymphadenopathy or splenomegaly; or have an active primary or recurrent malignancy; or have been in remission from a clinically significant malignancy for less than 5 years. a) Patients with cervical carcinoma in situ that has been resected without evidence of recurrence or metastatic disease for at least 3 years may participate in the study. b) Patients with basal cell or squamous cell skin cancer that has been completely removed without evidence of recurrence for at least 3 years may participate in the study. - Patients who have a current or recent, clinically serious viral, bacterial, fungal or parasitic infection, including Note: A recent viral infection of the upper respiratory tract or uncomplicated urinary tract should not be considered clinically serious. - Patients who have been exposed to a live vaccine within 12 weeks prior to the planned randomization date or are expected to need / receive a live vaccine during the course of the study (with the exception of shingles vaccination). - Have a history of chronic use of alcohol, injection drugs or other illicit drugs in the two years prior to screening. - Presence of a significant neuropsychiatric disorder that is uncontrolled, or clinically judged by the investigator to be at risk of suicide. - Have donated more than one unit of blood in the 4 weeks prior to screening or intend to donate blood during the study. |

(p. ex. asthme chronique instable). - Les patients qui ont été traités avec les traitements suivants: a) anticorps monoclonal (par exemple, ustekinumab, omalizumab, dupilumab) pendant moins de 5 demi-vies avant la randomisation. b) traitement préalable avec un inhibiteur oral de JAK (p. ex., tofacitinib, ruxolitinib) c) corticostéroïde systémique administré dans les 4 semaines précédant la randomisation planifiée ou devraient nécessiter des corticostéroïdes systémiques pendant l'étude. d) injection intra-articulaire de corticoïdes dans les 4 semaines précédant la date prévue de randomisation. e) plus de 250 séances de thérapies aux rayons UV. - Les patients qui sont dans l’incapacité totale ou partielle d’assurer leur propres soins (ex : alitement), comme être alités. - Les patients qui ont une hypertension artérielle non contrôlée caractérisée par une pression artérielle systolique répétée> 160 mm Hg ou une pression artérielle diastolique> 100 mm Hg en position assise. - Les patients qui ont subi une intervention chirurgicale majeure dans les 8 semaines précédant le screening ou qui nécessiteront une intervention chirurgicale majeure au cours de l'étude qui, de l'avis de l'investigateur, présenterait un risque inacceptable pour le patient. - Patients immunodéprimés et, de l'avis de l'investigateur, présentant un risque inacceptable de participer à l'étude. - Les patients qui ont présenté l'un des événements suivants dans les 12 semaines précédant la visite de sélection: événement thromboembolique veineux (TEV), infarctus du myocarde (IM), cardiopathie ischémique instable, accident vasculaire cérébral ou insuffisance cardiaque de stade III / IV de la NYHA. - Les patients qui ont des antécédents de TEV récidivant (≥ 2) ou qui sont considérés à haut risque de TEVselon l'investigateur. - Les patients ayant des antécédents ou présentant des troubles cardiovasculaires, respiratoires, hépatiques, gastro-intestinaux, endocriniens, hématologiques, neurologiques ou neuropsychiatriques ou toute autre maladie grave et / ou instable qui, de l'avis de l'investigateur, pourrait constituer un risque inacceptable avec la prise du produit expérimental ou interférer avec l'interprétation des données. - Les patients qui ont des antécédents de maladie lymphoproliférative; ou présentent des signes ou symptômes évocateurs d'une possible maladie lymphoproliférative, y compris des adénopathies ou une splénomégalie; ou ont une tumeur maligne primaire ou récurrente active; ou sont en rémission d'une tumeur maligne cliniquement significative depuis moins de 5 ans. a) Les patients atteints d'un carcinome cervical in situ qui a été réséqué sans signe de récidive ou de maladie métastatique pendant au moins 3 ans peuvent participer à l'étude. b) Les patients atteints de cancers cutanés basocellulaires ou épidermoïdes qui ont été complètement retirés sans signe de récidive depuis au moins 3 ans peuvent participer à l'étude. - Les patients qui ont une infection virale, bactérienne, fongique ou parasitaire actuelle ou récente, cliniquement grave, y compris Remarque: Une infection virale récente des voies respiratoires supérieures ou non compliquée des voies urinaires ne doit pas être considérée comme cliniquement grave. - Les patients qui ont été exposés à un vaccin vivant dans les 12 semaines précédant la date de randomisation prévue ou devraient avoir besoin / recevoir un vaccin vivant au cours de l'étude (à l'exception de la vaccination contre le zona). - Avoir des antécédents de consommation chronique d'alcool, de drogues injectables ou d'autres drogues illicites dans les deux années précédant le screening. - Présence d'un trouble neuropsychiatrique important non contrôlé, ou jugés cliniquement par l'investigateur comme étant à risque de suicide. - Avoir donné plus d'une unité de sang dans les 4 semaines précédant le screening ou avoir l'intention de donner son sang au cours de l'étude. |
