E.3 | Principal inclusion criteria |
To be eligible to the study, male and female subjects will have to fulfil the following criteria (assessment based on the medical examination performed at V0 with a checking at V1): I1. Age over 18 years (limit included), I2. Diagnosed with OIC according to the adapted ROME IV criteria (Mearin F et al, 2016) previously used in clinical evaluation of patients with OIC (Webster LR et al, 2017) and self-reported symptoms assessed by a physician (daily diary check at V1 visit): - Fewer than three (<3) spontaneous bowel movements (SBMs) per week. A SBM is defined as a bowel movement without the use of laxatives in the previous 24 hours as recorded in the daily diary. - And one or more of the following symptoms in at least 25% of bowel movements: straining, feeling of incomplete evacuation, and/or hard/small stools, defined as Bristol Stool Scale (BSS) score lower than 3 (<3). I3. With chronic non-cancer pain since at least three months, including the following indications but not limited to: back-pain, rheumatisms and post-operative pain, I4. Patients received opioids for chronic analgesia (intended treatment ≥6 Weeks) for 1 week or more prior to study start and a stable regimen of opioids for 3 or more days before study entry (V0 visit) and during the whole study , I5. Subjects must be willing to discontinue laxative use at screening and only use the rescue laxatives permitted throughout the study duration, I6. For women: - Non menopausal with the same reliable contraception since at least 2 cycles before the beginning of the study and agreeing to keep it during the entire duration of the study (condom with spermicide gel, any oral contraceptive, intrauterine device, subcutaneous contraceptive implant, vaginal ring, surgical intervention (bilateral tubal ligation or ovariectomy or hysterectomy), ESSURE system), - Menopausal without or with hormone replacement therapy, I7. Good general and mental health with in the opinion of the investigator: no clinically significant and relevant abnormalities of medical history or physical examination, I8. Able and willing to participate to the study by complying with the protocol procedures as evidenced by his dated and signed informed consent form, I9. Affiliated with a social security scheme.
Within 4 weeks following to the exit of the study for failure to comply with one or more of the inclusion criteria listed above, a re-screening could be performed. |
Les hommes et femmes, remplissant les critères suivants pourront-être inclus dans l’étude (évaluation sur la base de l'examen médical effectué à V0 et après vérification à V1) : I1. Agé de plus de 18 ans (limite incluse), I2. Diagnostiqué d’une CIO selon les critères adaptés de ROME IV (Mearin F et al, 2016) précédemment utilisés dans l'évaluation clinique des patients avec CIO (Webster LR et al, 2017) et symptômes auto-reportés évalués par un médecin (vérification du journal quotidien à la visite V1) : - Moins de trois (<3) selles spontanées (SBM) par semaine. Une SBM est définie comme une selle sans l'utilisation de laxatifs au cours des 24 heures précédentes, comme indiqué dans le journal quotidien. - Et un ou plusieurs des symptômes suivants dans au moins 25% des selles : effort, sensation d'évacuation incomplète et / ou selles dures / petites, définies par un score de Bristol Stool Scale (BSS) inférieur à 3 (<3). I3. Ayant une douleur chronique non cancéreuse depuis au moins trois mois, y compris les indications suivantes mais non limitées à : douleurs dorsales, rhumatismes et douleurs post-opératoires, I4. Patients ayant reçu des opioïdes pour une analgésie chronique (traitement prévu ≥ 6 semaines) pendant 1 semaine ou plus avant le début de l'étude et ayant une posologie stable d'opioïdes pendant 3 jours ou plus avant le début de l'étude (visite V0) et pendant toute l’étude, I5. Patients disposés à arrêter l'utilisation de laxatifs dès la visite V0 et à n'utiliser que les laxatifs de secours autorisés pendant toute la durée de l'étude, I6. Pour les femmes : - Non ménopausées avec la même contraception fiable depuis au moins 2 cycles avant le début de l'étude et acceptant de la conserver pendant toute la durée de l'étude (préservatif avec gel spermicide, tout contraceptif oral, dispositif intra-utérin, implant contraceptif sous-cutané, anneau vaginal, intervention chirurgicale (ligature des trompes ou ovariectomie bilatérale, hystérectomie, système Essure), - Ménopausées sans ou avec traitement hormonal substitutif,
I7. En bon état de santé général et mental selon l’avis de l’investigateur : pas d’anomalie cliniquement significative et pertinente selon les antécédents médicaux et l’examen clinique ; I8. Capable et désireux de participer à la recherche en se conformant aux procédures du protocole et en l’ayant signifié par la signature d’un formulaire de consentement éclairé daté ; I9. Affilié à un régime de sécurité sociale ;
Un re-screening peut être effectué à partir de 4 semaines près la sortie de l'étude pour non-conformité à l’un ou plusieurs des critères d’éligibilité énumérés plus haut. |

Subjects with the following criteria will be considered as non eligible to the study (assessment based on the medical examination performed at V0 and with a checking at V1): E1. Involvement in any investigational drug or device study within 30 days prior to this study, E2. Unable to withdraw other OIC treatments (any kind of laxative other than provided in the study), E3. History of chronic constipation prior to starting analgesic medication or any potential non-opioid cause of bowel dysfunction that may be a major contributor to the constipation upon investigator judgment, E4. Surgery planned within the whole study period, E5. Evidence of active medical diseases affecting bowel transit, E6. Antibiotic treatment intake within the last month prior the study start (V0), E7. Unwilling to withdraw probiotic supplements, yoghurts without supplemented bacteria are permitted, E8. Any history of drug addiction in the past five years, E9. Pregnant or lactating women or intending to become pregnant, E10. Unwilling to maintain food habits and current physical activity for the whole study duration, E11. With a personal history of anorexia nervosa, bulimia or significant eating disorders according to the investigator, E12. Having a lifestyle deemed incompatible with the study according to the investigator including drug and alcohol abuse, E13. Taking part in another clinical trial or being in the exclusion period of a previous clinical trial, E14. Under legal protection (guardianship, wardship) or deprived from his rights following administrative or judicial decision, E15. Presenting a psychological or linguistic incapability to sign the informed consent, E16. Impossible to contact in case of emergency.

Les patients avec les critères suivants ne seront pas éligibles à l'étude (évaluation sur la base de l'examen médical effectué à V0 et après vérification à V1) : E1. Incapable d’arrêter les autres traitements pour CIO (tout type de laxatif autre que celui fourni dans l'étude), E3. Ayant des antécédents de constipation chronique avant le début d'un traitement analgésique ou de toute cause potentielle non opioïde de dysfonctionnement intestinal qui pourrait être un contributeur majeur à la constipation selon l’avis de l'investigateur, E4. Ayant une chirurgie planifiée sur la période d'étude, E5. Preuve d’une maladie active affectant le transit intestinal, E6. Ayant consommé un traitement antibiotique sur le dernier mois avant le début de l'étude (V0), E7. Refusant d’arrêter sa consommation de suppléments probiotiques, les yaourts non supplémentés en bactéries sont autorisés, E8. Ayant un antécédent d’addiction de drogues au cours des cinq dernières années, E9. Femmes enceintes ou allaitantes ou ayant l'intention de devenir enceintes, E10. Non disposé à maintenir ses habitudes alimentaires et son activité physique actuelles pendant toute la durée de l'étude, E11. Ayant des antécédents personnels d'anorexie mentale, de boulimie ou de troubles alimentaires importants selon l'investigateur, E12. Ayant un mode de vie jugé incompatible avec l'étude selon l'investigateur incluant l'abus de drogues et d'alcool, E13.
