E.3 | Principal inclusion criteria |
Participants who have a diagnosis of CSU refractory to H1-AH at the time of randomization - Diagnosis of CSU >/= 3 months prior to screening visit (Visit 1). - The presence of itch and hives for >/= 6 consecutive weeks at any time prior to screening visit (Visit 1) despite the use of H1-AH during this time period. - Participants using a study defined H1-AH for CSU treatment. For participants on stable doses of non-study-approved H1-AH, investigators may switch participants to an equivalent dose of a study-approved H1- AH maintenance medication. - Participants who are omalizumab naïve. - Participants must be willing and able to complete a daily symptom e-diary for the duration of the study. - During the 7 days before randomization: UAS7 >/= 16 and ISS7 >/=8 - Body mass index (BMI) >17.5 and < 40 kg/m2. - Contraceptive use by men and women should be consistent with local regulations regarding the methods of contraception for those participating in clinical studies. |
Partecipanti che presentano una diagnosi di CSU refrattaria H1-AH al momento della randomizzazione - Diagnosi di CSU >/= 3 mesi prima della visita di screening (Visita 1). - Presenza di prurito e orticaria per >/=6 settimane consecutive in qualsiasi momento prima della visita di screening (Visita 1) nonostante l’uso di H1-AH durante questo periodo di tempo. - Partecipanti che utilizzano un H1-AH definito dallo studio per il trattamento della CSU. Per i/le partecipanti che assumono dosi stabili di H1-AH non approvate dallo studio, gli sperimentatori possono passare a una dose equivalente di un farmaco di mantenimento H1-AH approvato dallo studio. - Partecipanti naïve a omalizumab -I/Le partecipanti devono essere disposti/e e in grado di compilare un diario elettronico dei sintomi giornalieri per tutta la durata dello studio - Durante i 7 giorni precedenti la randomizzazione: UAS7 >/=16 e ISS7 >/=8 - Indice di massa corporea (IMC) >17,5 e < 40 kg/m2. - L’uso della contraccezione da parte di uomini e donne deve essere in linea con le normative locali riguardanti i metodi di contraccezione per coloro che partecipano agli studi clinici |

E.4 | Principal exclusion criteria |
Participants are excluded from the study if any of the following criteria apply: - Clearly defined underlying etiology for CUs other than CSU (main manifestation being physical urticaria) - Presence of skin morbidities other than CSU that may interfere with the assessment of the study outcomes. - Participants with active atopic dermatitis (AD). - Severe concomitant illness(es) that, in the Investigator's judgment, would adversely affect the patient's participation in the study. - Known or suspected immunodeficiency, or otherwise recurrent infections of abnormal frequency or prolonged duration suggesting an immune compromised status, as judged by the Investigator. - History of serious infections requiring intravenous (IV) therapy with the potential for recurrence or currently active moderate to severe infection at Screening, including active coronavirus disease 2019 (COVID-19). - Live vaccine except Bacille Calmette Guerin-vaccination within 28 days prior to Day 1 or plan to receive one during the trial; Bacille Calmette Guerin-vaccination within 12 months prior to Screening. - Active malignancy or history of malignancy within 5 years - Conditions that may predispose the participant to excessive bleeding - Any participant with an uncontrolled disease state as judged by the Investigator, such as asthma, psoriasis, or inflammatory bowel disease, etc. that are typically treated with oral or parenteral corticosteroids - Previous use of a BTK inhibitor. - Has received any investigational drug (or is currently using an investigational device) within the 30 days before Day 1, or at least 5 times the respective elimination half-life time (whichever is longer). - Previous exposure to another investigative antibody for CSU - Positive for human immunodeficiency virus (HIV) antibody test. - Presence of hepatitis B surface antigen (HBsAg) or hepatitis B core antibody (HBcAb) with positive DNA test result at screening or within 3 months prior to the screening visit. - Positive hepatitis C antibody test result at screening or within 3 months prior to the screening visit. - Tuberculosis infection - Any of significant laboratory abnormalities and ECG findings at the screening visit |
In presenza di uno dei seguenti criteri i/le partecipanti saranno esclusi dallo studio: - Eziologia sottostante chiaramente definita per CU diverse dalla CSU (la manifestazione principale è l’orticaria fisica). - Presenza di morbilità cutanee diverse da CSU che possono interferire con la valutazione degli esiti dello studio. - Partecipanti con dermatite atopica (DA) attiva. - Grave/i malattia/e concomitante/i che, a giudizio dello sperimentatore, possa/possano influire negativamente sulla partecipazione del/della paziente allo studio. - Anamnesi nota o sospetta di immunodeficienza o altrimenti infezioni ricorrenti insolitamente frequenti o di durata prolungata, che suggeriscono uno stato immunocompromesso, secondo il giudizio dello sperimentatore. - Anamnesi di infezioni serie che richiedono una terapia endovenosa (EV) con potenziale recidiva o infezione attualmente attiva da moderata a grave allo screening , compresa la malattia da coronavirus 2019 (COVID-19) - Vaccino vivo, a eccezione della vaccinazione con bacillo di Calmette-Guérin nei 28 giorni precedenti il Giorno 1 o previsione di riceverne uno durante la sperimentazione; vaccinazione con bacillo di Calmette-Guérin nei 12 mesi precedenti lo screening. - Malignità attiva o anamnesi di malignità nei 5 anni precedenti - Condizioni che possono predisporre il/la partecipante a sanguinamento eccessivo - Qualsiasi partecipante con uno stato di malattia non controllato in base al giudizio dello sperimentatore, come asma, psoriasi o malattia infiammatoria intestinale, ecc. che viene tipicamente trattato con corticosteroidi orali o parenterali. - Uso precedente di un inibitore di BTK. - Ha ricevuto qualsiasi farmaco sperimentale (o sta attualmente utilizzando un dispositivo sperimentale) nei 30 giorni precedenti il Giorno 1, o almeno 5 volte il rispettivo periodo di emivita di eliminazione (a seconda di quale sia il periodo più lungo). - Precedente esposizione a un altro anticorpo sperimentale per CSU. - Positività al test degli anticorpi contro il virus dell’immunodeficienza umana (HIV). - Presenza di antigene di superficie del virus dell’epatite B (HBsAg) o anticorpo anti-core dell’epatite B (HBcAb) con risultato positivo al test del DNA allo screening o nei 3 mesi precedenti la visita di screening. - Risultato positivo al test per gli anticorpi dell’epatite C allo screening o nei 3 mesi precedenti la visita di screening - Infezione da tubercolosi (TBI)
