E.3 | Principal inclusion criteria |
Male or female patients with age >= 18 years at screening,
Women of childbearing potential must be ready and able to use highly effective methods of birth control,
Have a diagnosis of chronic plaque psoriasis (with or without psoriatic arthritis) for at least 6 months before the first administration of study drug.
Have stable moderate to severe chronic plaque psoriasis with or without psoriatic arthritis at both Screening and Baseline (Randomization): a. Have an involved body surface area (BSA) >= 10% and b. Have a Psoriasis Area and Severity Index (PASI) score >= 12 and c. Have a static Physician Global Assessment (sPGA) score of >= 3.
Must be candidates for systemic therapy or phototherapy for psoriasis treatment, as assessed by the investigator
Must be a candidate for treatment with Stelara® (ustekinumab) according to local label. |
-Maschi o femmine. Le femmine potenzialmente fertili devono essere disponibili e capaci di usare un metodo contraccettivo altamente efficace secondo le linee guida ICH M3 (R2), che risulta in un basso tasso di fallimento, ad esempio meno dell’1% annuo, quando usato coerentemente e correttamente (la lista di tali metodi contraccetivi sarà inclusa nell’informativa paziente). -Età di almeno18 anni alla visita di screening. -Diagnosi di psoriasi cronica a placche (con o senza artrite psoriasica) accertata almeno sei mesi prima della prima somministrazione del farmaco in studio. La durata della diagnosi può essere riportata dal paziente. -Psoriasi cronica a placche stabile da moderata a grave con o senza artrite psoriasica sia allo screening che alla visita basale (randomizzazione): -BSA >=10% e -PASI >= 12 e -sPGA >= 3 -Candidato per la terapia sistemica o la fototerapia per il trattamento della psoriasi in base al giudizio dello sperimentatore. -Candidato al trattamento con Stelara (ustekinumab) in conformità all’indicazione locale. -Consenso informato firmato e datato in conformità alle GCP e alle normative locali prima della partecipazione alla sperimentazione. |

Patients with: a. non-plaque forms of psoriasis (including guttate, erythrodermic, or pustular), b. current drug-induced psoriasis (including an exacerbation of psoriasis from beta blockers, calcium channel blockers, or lithium), c. active ongoing inflammatory diseases other than psoriasis and psoriatic arthritis that might confound trial evaluations according to investigators judgment, Previous exposure to BI 655066(risankinumab) , Previous exposure to ustekinumab (Stelara®), Currently enrolled in another investigational study or less than 30 days (from screening) since completing another investigational study (participation in observational studies is permitted), Use of any restricted medication , or any drug considered likely to interfere with the safe conduct of the study, Major surgery performed within 12 weeks prior to randomization or planned within 12 months after screening (e.g. hip replacement, aneurysm removal, stomach ligation), Known chronic or relevant acute infections including active tuberculosis, HIV or viral hepatitis, Any documented active or suspected malignancy or history of malignancy within 5 years prior to screening, except appropriately treated basal or squamous cell carcinoma of the skin or in situ carcinoma of uterine cervix, Evidence of a current or previous disease, medical condition (including chronic alcohol or drug abuse) other than psoriasis, surgical procedure (i.e., organ transplant), medical examination finding (including vital signs and ECG), or laboratory value at the screening visit outside the reference range that is in the opinion of the investigator, is clinically significant and would make the study participant unreliable to adhere to the protocol or to complete the trial, compromise the safety of the patient, or compromise the quality of the data, History of allergy/hypersensitivity to a systemically administered biologic agent or its excipients, Women who is pregnant, nursing, or who plans to become pregnant while in the trial, -Subjects who were exposed to live vaccines in the 6 weeks prior to randomization |
-Pazienti con forme di psoriasi non a placche (compreso guttata , eritrodermica o pustolosa); psoriasi attiva indotta da farmaci (tra cui una esacerbazione della psoriasi da beta-bloccanti , calcio-antagonisti, o litio); malattie infiammatorie attive in corso diverse da psoriasi e artrite psoriasica che potrebbero confondere le valutazioni dello studio in base al giudizio dello sperimentatore. -Precedente esposizione a BI 655066 (risankinumab). -Pazienti che abbiano partecipato ad un altro studio con un farmaco o dispositivo sperimentale più o meno da 30 giorni precedenti la visita di screening o che stiano partecipando ad un altro studio (la partecipazione a studi osservazionali è consentita). -Precedente esposizione ad ustekinumab (Stelara). -Uso di qualsiasi farmaco concomitante come specificato nella tabella sotto riportata, o qualsiasi farmaco che possa interferire con lo svolgimento sicuro dello studio. -Chirurgia maggiore eseguita entro 12 settimane precedenti la randomizzazione o pianificata entro 12 mesi dopo lo screening (per esempio protesi d'anca, la rimozione aneurisma, legatura stomaco). -Infezioni croniche o acute rilevanti note, tra cui tubercolosi attiva , HIV o epatiti virali; il test QuantiFERON TB o test cutaneo PPD verranno eseguiti secondo l'etichettatura locale per Stelara. Se il risultato è positivo, i pazienti possono partecipare allo studio se ulteriori controlli medici (secondo le prassi e linee guida locali) stabiliscono definitivamente che il paziente non ha evidenze di tubercolosi attiva . Se viene stabilita la presenza di tubercolosi latente, quindi il trattamento avrebbe dovuto essere iniziato e mantenuto secondo le linee guida nazionali locali. -Presenza documentata o sospetta presenza di tumore maligno attivo o passato entro i 5 anni prima dello screening, ad eccezione del carcinoma a cellule squamose o basali della pelle o del carcinoma in situ della cervice uterina opportunamente trattati. -Attiva o pregressa malattia, condizione medica (incluso l’abuso cronico di alcool e droghe) diverse dalla psoriasi, procedura chirurgica (ad esempio trapianto di organo), evidenze dell’esame obiettivo (incluso segni vitali ed ECG) o valori di laboratorio fuori dal range di riferimento alla visita di screening che, secondo lo sperimentatore, sono clinicamente significative e potrebbero mettere il partecipante non in condizioni di aderire alle procedure dello studio, mettere a rischio la sicurezza del paziente oppure compromettere la qualità dei dati. -Storia di allergia o ipersensibilità ad un agente biologico somministrato per via sistemica o dei suoi eccipienti. -Donne in gravidanza, che allattano o che hanno pianificato una gravidanza durante il periodo di partecipazione allo studio. -Precedente arruolamento in questo studio. -Pazienti sottoposti a trattamento con vaccini vivi nelle 6 settimane prima della randomizzazione |
