1. Age (at the time of consent) ≥18 years of age 2. Has an Eastern Cooperative Oncology Group (ECOG) performance status of 0 or 1 3. Has a life expectancy of >3 months 4.Has mesothelioma (pleural, peritoneal, pericardial, tunica vaginalis) of any histology that is relapsed or refractory after treatment with at least one pemetrexed-containing regimen 5.Has a documented local diagnostic pathology of original biopsy confirmed by a Clinical Laboratory Improvement Amendments (CLIA)/College of American Pathologists (CAP) or equivalent laboratory certification 6.Part 2: Molecular evidence of BAP1 loss of function mutation present on local pathology, e.g., lack of nuclear BAP1 staining by immunohistochemistry (IHC) or evidence of loss of function by gene sequencing 7. Has sufficient archival tumor tissue (a minimum of 10 slides or tumor block) available for central retrospective testing of BAP1 status 8. Has all prior treatment (i.e., chemotherapy, immunotherapy, radiotherapy) related clinically significant toxicities resolve to ≤ Grade 1 per CTCAE, version 4.03 or are clinically stable and not clinically significant, at time of enrollment 9. Prior therapy(ies), if applicable, must be completed according to the criteria below prior to first dose of tazemetostat: a. Cytotoxic chemotherapy; at least 21 days since last dose b. Non-cytotoxic chemotherapy (e.g., small molecule inhibitor); at least 14 days since last dose c. Monoclonal antibody; at least three half-lives since the last dose d. Non-antibody immunotherapy (e.g., tumor vaccine); at least 42 days since last dose e. Radiotherapy, at least 14 days from last local site radiotherapy f. Hematopoietic growth factor; at least 14 days from last dose g. Investigational drug; 30 days or five half-lives, whichever is longer, from last dose 10. Has measurable disease based on either modified RECIST [Nowak 2005] for thoracic disease or RECIST 1.1 elsewhere 11. Has adequate hematologic (bone marrow and coagulation factors), renal, and hepatic function as defined by criteria below: a. Hemoglobin ≥9 mg/dL b. Platelets ≥100,000/mm3 (≥100 × 109/L) without platelet transfusion for 7 days c. ANC ≥1000/mm3 (≥1.0 × 109/L) without growth factor support for 14 days d. Coagulation: Prothrombin time (PT) <1.5 × ULN and partial thromboplastin time (PTT) <1.5 × ULN e. Creatinine < 2.0 × ULN f. Hepatic function: Conjugated bilirubin <1.5 × ULN and ALT and AST <3 × ULN 12. Has a QT interval corrected by Fridericia's formula (QTcF) ≤480 msec 13. Female subjects of childbearing potential must have a negative urine or serum pregnancy within 72 hours prior to receiving the first dose of study drug; if the urine test is positive or cannot be confirmed as negative, a serum pregnancy test will be required and subject also should agree to use an adequate method of contraception starting with screening through 30 days after the last dose of study therapy (if sexually active). 15. Avoir un examen anatomo-pathologique local documenté de la biopsie originale confirmé par un laboratoire certifié par le CLIA (Clinical Laboratory Improvement Amendments)/CAP (College of American Pathologists) ou certification équivalente 6. 2ème partie : présence d'une preuve moléculaire de mutation perte de fonction BAP1 en pathologie locale, par ex. manque de marquage BAP1 nucléaire par immunohistochimie (IHC) ou confirmation de la perte de fonction par séquençage génétique 7. Disposer de suffisamment de tissu tumoral d'archivage (au moins 10 lames ou un bloc tumoral) pour une analyse rétrospective centrale du statut BAP1 8. Toutes les toxicités cliniquement significatives liées aux traitements précédents (chimiothérapie, immunothérapie, radiothérapie) ont été ramenées au ≤ grade 1, conformément à la classification CTCAE, version 4.03, ou sont cliniquement stables et non cliniquement significatives à la date du recrutement 9. Le cas échéant, le ou les traitements antérieurs à la première administration de tazemetostat doivent être achevés, conformément aux critères ci-dessous: a. Radiothérapie : au moins 14 jours depuis la dernière séance de radiothérapie locale f. Facteur de croissance hématopoïétique : au moins 14 jours depuis la dernière dose g. Avoir une maladie mesurable selon les critères RECIST modifiés [Nowak, 2005] pour les maladies thoraciques ou RECIST 1.1 pour les autres régions du corps 11. Présenter une fonction hématologique (moelle osseuse et facteurs de coagulation), rénale et hépatique adéquate telle que définie par les critères ci-dessous : a. Hémoglobine ≥ 9 mg/dl b. Plaquettes ≥ 100 000/mm3 (≥ 100 × 109/l) sans transfusion de plaquettes pendant 7 jours c. NAN ≥ 1 000/mm3 (≥ 1,0 × 109/l) sans traitement par facteur de croissance pendant 14 jours d. Coagulation : temps de prothrombine (TP) < 1,5 × LSN et temps de thromboplastine partielle (TTP) < 1,5 × LSN e. Créatinine < 2,0 × LSN f. Fonction hépatique : bilirubine conjuguée < 1,5 × LSN et ALAT et ASAT <3 × LSN 12. Avoir un intervalle QT corrigé par la formule de Fridericia (QTcF) ≤ 480 ms 13. Consentir à l'utilisation des tissus pour la recherche translationnelle 14. Les patientes de sexe féminin en âge de procréer doivent présenter un test de grossesse urinaire ou sérique négatif dans les 72 heures précédant la prise de la première dose du médicament à l'étude. Si le test urinaire est positif ou n'est pas concluant (négativité douteuse), un test de grossesse sérique sera exigé et la patiente doit aussi accepter d'utiliser une méthode de contraception adéquate à compter de sa sélection et jusqu'à 30 jours après la prise de la dernière dose du traitement à l'étude (si elle est sexuellement active). 15. Les patients de sexe masculin doivent accepter d'utiliser des préservatifs à compter de la prise de la première dose du traitement à l'étude et jusqu'à 30 jours après la prise de la dernière dose s'ils sont sexuellement actifs et ont pour partenaire une femme en âge de procréer.

1. Has had prior exposure to tazemetostat or other inhibitor(s) of enhancer of zeste homologue-2 (EZH2) 2. Has a history of known central nervous system metastasis 3. Has had a prior malignancy other than the malignancies under study Exception: A subject who has been disease-free for 5 years, or a subject with a history of a completely resected non-melanoma skin cancer or successfully treated in situ carcinoma is eligible. 4. Has had major surgery within 3 weeks prior to enrollment (a percutaneous biopsy, pleural catheter insertion, placement of central venous catheter or other minor procedure are permitted) 5. Is unwilling to exclude grapefruit juice, Seville oranges and grapefruit from the diet and all foods that contain those fruits from time of enrollment throughout their time on study 6. Has cardiovascular impairment, history of congestive heart failure greater than NYHA Class II, uncontrolled arterial hypertension, unstable angina, myocardial infarction, or stroke within 6 months prior to the planned first dose of tazemetostat; or ventricular cardiac arrhythmia requiring medical treatment 7. Is currently taking any prohibited medication(s) 8. Has an active infection requiring systemic treatment 9. Has a congenital or acquired immunodeficiency, including subjects with known history of infection with human immunodeficiency virus (HIV) NOTE: HIV-positive subjects who are taking antiretroviral therapy are ineligible due to potential PK interactions with tazemetostat. 10. Has known history of chronic infection with hepatitis B virus (hepatitis B surface antigen positive) or hepatitis C virus (detectable anti-hepatitis C circulating viral RNA) 11. Has had a deep venous thrombosis (DVT) or pulmonary embolism within the 3 months prior to study enrollment. NOTE: Subjects with a history of a DVT or pulmonary embolism >3 months prior to study enrollment who are on anticoagulation therapy with low molecular weight heparin are eligible for this study. 12. Is pregnant or breastfeeding |
1. Avoir été précédemment exposé au tazemetostat ou à un ou plusieurs autres inhibiteurs de l'améliorateur de l'homologue 2 de zeste (EZH2) 2. Avoir des antécédents connus de métastase du système nerveux central 3. Avoir déjà souffert d'une affection maligne autre que les affections malignes à l'étude Exception : un patient en rémission depuis 5 ans, ou avec des antécédents de cancer de la peau autre qu'un mélanome ayant fait l'objet d'une résection totale ou de carcinome in situ ayant été traité avec succès est éligible 4. Avoir subi une intervention chirurgicale majeure dans les 3 semaines précédant le recrutement (la biopsie percutanée, la mise en place d'un cathéter pleural, la mise en place d'un cathéter veineux central ou toute autre intervention mineure sont autorisées) 5. Refuser d'exclure le jus de pamplemousse, les oranges amères et le pamplemousse de son régime alimentaire, ainsi que tous les aliments contenant ces fruits à compter de la date de recrutement et pendant toute la participation à l'étude 6. Avoir présenté une défaillance cardiovasculaire, des antécédents d’insuffisance cardiaque congestive d'une sévérité supérieure à la classe II de la NYHA, une hypertension artérielle non contrôlée, un angor instable, un infarctus du myocarde, ou un AVC au cours des 6 mois précédant la première prise prévue de tazemetostat ; ou une arythmie cardiaque ventriculaire nécessitant un traitement médical 7. Prendre actuellement un ou plusieurs médicaments interdits 8. Avoir une infection active nécessitant un traitement systémique 9. Souffrir d'immunodéficience congénitale ou acquise, y compris les patients ayant des antécédents connus d'infection par le virus d'immunodéficience humain (VIH) REMARQUE : les patients positifs au VIH qui prennent des antirétroviraux sont inéligibles à cause des interactions PK potentielles avec le tazemetostat. 10.
