E.3 | Principal inclusion criteria |
1. Participant is >= 60 to <= 90 years of age at the time of signing the informed consent. 2. Participant is an ambulatory postmenopausal woman, diagnosed with osteoporosis, able to walk and not bedridden. A woman is considered postmenopausal if she meets any of the following criteria: a. Has 12 months of spontaneous amenorrhea without an alternative medical cause with serum follicle stimulating hormone (FSH) levels falling in the normal postmenopausal range at the central laboratory at the time of the Screening Period. b. Female participant underwent bilateral oophorectomy (with or without hysterectomy) at least 6 weeks prior to the Screening Period. 3. Participant has an absolute BMD consistent with T score <= 2.5 and >= 4.0 at the lumbar spine as measured by dual energy X ray absorptiometry (DXA) during the Screening Period and at least 2 lumbar vertebrae (from L1 to L4) must be evaluable by DXA. 4. Participant has body weight >= 50 and <= 90 kg at the Screening Period. 5. Participant is willing and able to give signed informed consent which includes compliance with the requirements and restrictions listed in the informed consent form (ICF) and in the protocol. 6. Participant has provided informed consent prior to any study specific activities/procedures.
Participant must meet the following criteria to be enrolled in the Transition Period: 7. Have been enrolled, received both doses of IMP and completed the scheduled Main Period (up to Week 52) of the RGB-14-101 study. |
1. La partecipante ha un’eta >= 60 e <= 90 anni al momento della firma del consenso informato. 2. La partecipante è una donna in menopausa ambulatoriale, con diagnosi di osteoporosi, in grado di camminare e non costretta a letto. Una donna è considerata in menopausase soddisfa uno dei seguenti criteri: a. Presenta 12 mesi di amenorrea spontanea senza una causa medica alternativa con livelli sierici dell’ormone follico stimolante (FSH) che rientra nel normale intervallo postmenopausale presso il laboratorio centralizzato al momento del periodo di screening. b. la partecipante è stata sottoposta a ovariectomia bilaterale (con o senza isterectomia) lameno 6 settimane prima del periodo di screening 3. La partecipante ha un BMD assoluta coerente con un punteggio T <= 2,5 e >= 4,0 a livello della colonna lombare misurata mediante assorbimetria a raggi X a doppia energia (DXA) durante il periodo di screening e almeno 2 vertebre lombari (dalla L1 alla L4) devono essere valutabili dal DXA. 4. La partecipante ha un peso corporeo >= 50 e <= 90 Kg al periodo di screening. 5. La partecipante è disposta e in grado di fornire un consenso informato firmato che includa la conformità con i requisiti e le restrizioni elencate nel modulo di consenso informato (ICF) e nel protocollo. 6. La partecipante ha fornito il consenso informato prima di qualsiasi attività/procedura specifica dello studio.

E.4 | Principal exclusion criteria |
1. Participant has a history and/or presence of one severe or more than two moderate vertebral fractures as determined by central reading of lateral spine X ray during the Screening Period. 2. Participant has a history and/or presence of hip fracture. 3. Participant has a history and/or presence of atypical femur fracture. 4. Participant presents with an active healing fracture according to the assessment of the Investigator. 5. Participant has a bilateral hip replacement (unilateral is allowed if the other hip is evaluable with DXA). 6. Participant has a vitamin D deficiency, the assessment may be repeated once after vitamin D supplementation. Appropriate vitamin D dosing in addition to vitamin D supplementation is permitted at the discretion of the Investigator during the Screening Period. 7. Participant has hypocalcaemia or hypercalcaemia at the Screening Period, the assessment may be repeated once in the case of hypercalcaemia. 8. Participant has a history and/or presence of bone metastases, renal osteodystrophy, osteomyelitis, any metabolic, endocrine or traumatic bone disease (except postmenopausal osteoporosis) that may interfere with the interpretation of the results. 9. Participant has a current uncontrolled status of hypothyroidism or hyperthyroidism. 10. Participant has a history (within 5 years prior to Screening) and/or current hypoparathyroidism or hyperparathyroidism other than clinically not significant secondary hyperparathyroidism. 11. Participant has had major surgery within 8 weeks prior to the Screening Period or planned, anticipated major surgery during the study. 12. Participant has malignancy within 5 years before Screening (except completely cured in situ cervical carcinoma or non metastatic squamous or basal cell carcinoma of the skin). 13. Participant has a history and/or presence of significant cardiac disease or ECG abnormalities indicating significant risk for participating in the study as judged by the Investigator. 14. Participant has a known intolerance or malabsorption of calcium or vitamin D supplements. 15. Participant has a known hypersensitivity (including severe allergic reaction) to monoclonal antibodies or a history of systemic hypersensitivity to any components of the IMPs. 16. Participant shows contraindications to denosumab therapy (e.g., hypocalcaemia), or calcium or vitamin D supplementation before starting IMP administration. 17. Participant has a latex allergy. 18. Participant has a history and/or presence of ONJ or risk factors for ONJ. 19. Participant has history and/or presence of osteonecrosis of the external auditory canal. 20. Participant requiring ongoing use of any osteoporosis treatment (other than calcium and vitamin D supplements). 21. Participant has previously received denosumab or biosimilar denosumab. 22. Participant has previously received any of medicines listed in protocol. 23. Participant is receiving or has received any investigational drug (or is currently using an investigational device) within 3 months before receiving IMP, or at least 10 times the respective elimination half life (whichever period is longer). 24. Participant has weight or girth measurements which may preclude accurate DXA measurements in Investigator's opinion.
For all the other criteria, refer to the Protocol
Participant meeting the following criteria must not be enrolled in the Transition Period: 30. Treatment continuation in the study is against the best interest of the participant as deemed by the Investigator. |
1. La partecipante ha un passato e/o la presenza di una o più di due fratture vertebrali moderate come determinato dalla lettura centralizzata della radiografia della colonna vertebrale laterale durante il periodo di screening. 2. La partecipante ha un passato e/o la presenza di frattura dell’anca. 3. La partecipante ha un passato e/o la presenta di frattura al femore atipica. 4. La partecipante si presenta con una fattura in via di guarigione attiva secondo la valutazione dello sperimentatore 5. La partecipante ha una protesi all’anca bilaterale (unilaterale è consentito se l’altra anca è valutabile con DXA). 6. La partecipante ha una carenza di vitamina D, la valutazione può essere ripetuta una volta dopo l’integrazione della vitamina D. Un dosaggio appropriato di vitamina D in aggiunta all’integrazione della vitamina D è consentito a discrezione dello sperimentatore durante il periodo di screening. 7. La partecipante ha ipocalcemia o ipercalcemia durante il periodo di screening, la valutazione può essere ripetuta una volta in caso di ipercalcemia. 8. La partecipante ha un passato e/o la presenza di metastai ossee, osteodistrofia renale, osteomielite, qualsiasi malattia ossea metabolica, endocrina o tramatica (eccetto l’osteoporosi postmenopausale) che può interferire con l’interpretazione dei risultati. 9. La partecipante ha uno stato attuale non controlllato di ipotiroidismo o ipertiroidismo. 10. La partecipante ha un passato (nei 5 anni prima dello screening) e/o un ipoparatiroidismo o iperparatiroidismo in corso oltre all’iperparatiroidismo secondario clinicamente non significativo. 11. La partecipante ha subito un intervento chirurgico importante nelle 8 settimane precedenti il periodo di screening o un intervento chirurgico importante pianificato, anticipato durante lo studio. 12. La partecipante ha avuto un tumore maligno nei 5 anni precedenti lo screening (eccetto il carcinoma cervicale in situ completamente curato o il carcinoma a cellule basali o squamose non metastatico della pelle). 13. La partecipante ha un passato e/o la presenza di malattie cardiache significative o anomalie dell’ECG che indicano un rischio significativo per la partecipazione allo studio secondo il giudizio dello sperimentatore. 14. La partecipante ha un’intolleranza nota o un malassorbimento di integratori di calcio o vitamina D. 15. La partecipante ha una nota ipersensibilità (inclusa una grave reazione allergica) agli anticorpi monoclonali o un passato di ipersensibilità sistemica a qualsiasi componente degli IMP. 16. La partecipante mostra controindicazioni alla terapia con denosumab (ad es. Ipocalcemia) o all’integrazione di calcio o vitamina D prima di iniziare la somministrazione degli IMP. 17. La partecipante ha un’allergia al lattice. 18. La partecipante ha un passato e/o la presenza di ONJ o fattori di rischio per ONJ. 19. La partecipante ha un passato e/o la presenza di osteonecrosi del canale uditivo esterno. 20. Partecipante che richiede l’uso continuo di qualsiasi trattamento per l’osteoporosi (diversi dagli integratori di calcio e vitamina D). 21. La partecipante ha già ricevuto denosumab o denosumab biosimilare. 22. La partecipante ha ricevuto in precedenza uno qualsiasi dei medicinali elencati nel protocollo. 23. La partecipante riceve o ha ricevuto un farmaco sperimentale (o sta attualmente utilizzando un dispositivo sperimentale) entro 3 mesi prima di ricevere l'IMP, o almeno 10 volte la rispettiva emivita di eliminazione (a seconda di quale periodo è più lungo). 24. La partecipante ha misure di peso o circonferenza che, secondo lo sperimentatore, possono precludere misurazioni accurate della DXA.
